益气养阴方对糖尿病性脑内微小病变实验大鼠的保护作用

Protective Effect of Yin-Nourishing Therapy of Traditional Chinese Medicine on Experimental Rats with Diabetic Encephalopathy

目的 观察益气养阴方对糖尿病性脑内微小病变大鼠治疗效果。方法 建立大鼠糖尿病性脑内微小病变模型,将动物分为正常对照组、高糖损伤组、阿司匹林对照组、中药组,通过生化、流式细胞仪(PI)测定等方法检测大鼠脑内微小病变的变化;通过Real time PCR、Western blot测定HIF-1α的变化。结果 糖化血红蛋白及流式细胞结果提示,与高糖损伤组相比,阿司匹林组、中药组糖化血红蛋白、凋亡率都有所减少,且各组之间相比有明显统计学差异(P<0.05)。与阿司匹林组相比,中药组糖化血红蛋白表达量有所增加(P<0.05)。与阿司匹林组相比,中药组早期凋亡率所增加,但无明显统计学意义(P>0.05);中药组晚期凋亡率所增加且有统计学意义(P<0.05);与阿司匹林组相比,中药组HIF-1α mRNA及蛋白表达量有所增加(P<0.05)。Real time PCR、Western blot测定HIF-1α的变化发现,与高糖损伤组相比,阿司匹林、中药组、HIF-1α mRNA及蛋白表达量都有所减少(P<0.05),且各组之间相比有明显统计学差异(P<0.05),与阿司匹林组相比,中药组HIF-1α mRNA及蛋白表达量有所增加(P<0.05)。结论 益气养阴方对糖尿病性脑内微小病变的治疗具有一定的作用,可降低糖化血红蛋白吸光度,降低脑神经细胞凋亡率,降低HIF-1α的表达,相对于阿司匹林的治疗作用有一定差别。

Objective To observe the therapeutic effect of Yiqi Yangyin Recipe on diabetic intracerebral microlesions in rats.Methods The model of diabetic intracerebral microlesions in rats was established.The animals were divided into normal control group,high glucose injury group,aspirin control group and traditional Chinese medicine group.The changes of intracerebral microlesions in rats were detected by biochemistry and flow cytometry(PI),and the changes of HIF-lαwere detected by Realtime PCR and Western blot.Results The results of glycated hemoglobin and flow cytometry showed that compared with the high glucose injury group,the glycated hemoglobin and apoptotic rate of aspirin group and traditional Chinese medicine group decreased,and there was significant difference among the groups(P<0.05).Compared with aspirin group,the expression of glycosylated hemoglobin in traditional Chinese medicine group increased(P<0.05).Compared with aspirin group,the early apoptotic rate of Chinese herbal medicine group increased,but there was no significant difference(P>0.05);the late apoptotic rate of Chinese herbal medicine group increased with statistical significance(P<0.05);compared with aspirin group,the expression of HIF-1αin Chinese herbal medicine group increased(P<0.05).Realime PCR and Western blot showed that the expression of HIF-lαin aspirin,traditional Chinese medicine and HIF-lαdecreased compared with the high-glucose injury group(P<0.05),and there were significant dfferences among the groups(P<0.05).Compared with aspirin group,the expression of HIF-1αin traditional Chinese medicine group increased(P<0.05).Conclusion Yiqi Yangyin Recipe has a certain effect on the treatment of diabetic encephalopathy.It can reduce the absorbance of glycosylated hemoglobin,the apoptotic rate of brain neurons and the expression of HIF-lalpha.The therapeutic effect of aspirin is different from that of aspirin.
