半夏白术天麻汤“异病同治”高血压、高脂血症、眩晕的网络药理学研究

Network Pharmacologic Study of Banxia Baizhu Tianma Decoction in the Treatment of Hypertension,Hyperlipidemia and Vertigo by "Treating Different Diseases with Same Method"

目的:基于网络药理学探讨半夏白术天麻汤“异病同治”高血压、高脂血症、眩晕的作用机制。方法:通过TCMSP、BATMAN-TCM、SwissTargetPrediction、GeneCards数据库获取药物靶点和疾病靶点,通过STRING数据库和Cytoscape软件绘制蛋白-蛋白互作(PPI)网络图,利用Metascape数据库进行基因富集分析,利用AutoDock软件进行分子对接。结果:共检索到半夏白术天麻汤药物靶点847个、“异病同治”靶点65个。关键靶点为清蛋白(ALB)、丝氨酸/苏氨酸激酶1(AKT1)、白细胞介素6(IL6)、肿瘤坏死因子(TNF)、血管内皮生长因子A(VEGFA);关键成分为卡文定碱、黄甘草异黄酮A、柚皮素、甘草查尔酮B、山柰酚、黄芩素;关键靶点主要作用于肝、心、视网膜、松果体、甲状腺等器官组织,主要富集于循环系统的调控、蛋白磷酸化正调控、细胞钙离子稳态、血压负调节等生物过程及缺氧诱导因子-1(HIF-1)、辅助性T细胞17(Th17)细胞分化、环磷酸腺苷(cAMP)、流体剪切应力与动脉粥样硬化、过氧化物酶体增殖物激活受体(PPAR)等信号通路。分子对接结果显示配体和受体结合稳定。结论:半夏白术天麻汤“异病同治”高血压、高脂血症、眩晕的机制可能为关键成分通过多靶点、多通路,发挥抑制炎症、降低血脂、扩张血管等作用。

Objective:To explore the mechanism of Banxia Baizhu Tianma Decoction in treating hypertension,hyperlipidemia,and vertigo by "treating different diseases with same method" based on network pharmacology.Methods:Drug targets and diseases target were obtained by TCMSP,BATMAN-TCM,SwissTargetPrediction and GeneCards database.Protein-protein interaction(PPI) diagram was constructed by STRING database and Cytoscape software.Metascape database was used for gene enrichment analysis,and AutoDock software was used for molecular docking.Results:A total of 847 drug targets of Banxia Baizhu Tianma Decoction and 65 targets of " treating different diseases with same method " were retrieved.The key targets were albumin(ALB),serine/threonine kinase 1(AKT1),interleukin 6(IL6),tumor necrosis factor(TNF),vascular endothelial growth factor A(VEGFA),and the key components were Cavidine,Eurycarpin A,Naringenin,Licochalcone B,Kaempferol,and Baicalein.The key targets mainly acted on liver,heart,retina,pineal gland,thyroid gland,and other organs and tissues.The key targets were mainly focused on the regulation of circulatory system,positive regulation of protein phosphorylation,cellular calcium homeostasis,negative regulation of blood pressure and other biological processes,hypoxia-inducing factor-1(HIF-1) pathway,differentiation of T cell 17(Th17) cells,cyclic adenosine phosphate(cAMP) pathway,fluid shear stress and atherosclerosis,peroxisome proliferator activated receptors(PPAR) pathway and other signaling pathways.Molecular docking results showed that the ligand and receptor were stable.Conclusion:The mechanism of Banxia Baizhu Tianma Decoction in treating hypertension,hyperlipidemia,and vertigo by "treating different diseases with same method" might be related the special key ingredient which played some role of inhibiting inflammation,reducing blood lipid,and dilating blood vessels by acting as multiple targets and multiple pathways.
