电子烟对气道Clara细胞功能损害的研究

摘要 目的探索不同浓度电子烟对气道Clara细胞功能的损伤和调节炎症功能的影响。方法探查对照组0%,干预组4%、8%、12%、16%、20%、50%浓度的电子烟雾提取物(E-CSE)对Clara细胞12、24、48 h的损伤情况,探索最佳刺激浓度和时间。ELISA法分别检测对照组和干预组不同浓度的电子烟雾提取物刺激Clara细胞上清液CC16的含量。ELISA法分别检测对照组和干预组不同浓度的E-CSE干预Clara细胞后炎症因子白介素-1β(IL-1β)的含量。结果CCK-8法检测提示,不同浓度电子烟雾提取物干预组对Clara细胞12 h的抑制率均高于对照组,差异有统计学意义(P<0.05),浓度越高对Clara细胞抑制率越高。8%、12%、16%浓度E-CSE组Clara细胞12 h后CC16表达量低于对照组,差异有统计学意义(P<0.05)。12%、16%浓度E-CSE组Clara细胞IL-1β的含量均高于对照组,差异有统计学意义(P<0.05)。8%浓度E-CSE浓度含量与对照组比较,差异无统计学意义(P>0.05)。结论电子烟雾提取物会对Clara细胞及CC16产生抑制,而且随着浓度增大,抑制率明显增大。电子烟雾可影响Clara细胞炎症介质分泌,并且电子烟雾浓度越大,Clara细胞炎症介质分泌增多。 Objective To explore the effects of different concentrations of electronic cigarettes on the function injury and inflammation regulation of airway Clara cells.Methods The damage of electronic cigarette extract(E-CSE)with concentration of 0%in control group and 4%,8%,12%,16%,20%and 50%in intervention group on Clara cells for 12,24 and 48 hours were investigated to explore the optimal stimulation concentration and time.ELISA was used to ascertain the CC16 content of Clara cell supernatant,stimulated by electron smog extract in both the control and intervention groups.The ELISA technique was employed to ascertain the interleukin-1β(IL-1β)content post-intervention of Clara cells with varying levels of E-CSE in both the control and intervention groups.Results CCK-8 method indicated that the inhibition rate of 12 h of Clara cells in the intervention group with different concentrations of electronic smog extract was higher than that in the control group,and the differences were statistically significant(P<0.05).The higher the concentration was,the higher the inhibition rate of Clara cells was.The expression levels of CC16 in Clara cells in 8%,12%and 16%E-CSE groups after 12 h were lower than that in control group,and the differences were statistically significant(P<0.05).The content of IL-1βin Clara cells in 12%and 16%E-CSE groups were higher than that in control group,and the differences were statistically significant(P<0.05).There was no significant difference in concentration in 8%E-CSE group compared with control group(P>0.05).Conclusion Electronic smog extract can inhibit Clara cells and CC16,and the inhibition rate increases with the increase of concentration.Electron smog could affect the secretion of inflammatory mediators in Clara cells,and the secretion of inflammatory mediators in Clara cells increased with the concentration of electronic smog.
