衍生肽Esc-1GN(1-24)能减轻细胞氧化应激损伤和调节细胞因子抑制炎症反应

摘要 目的 探究衍生肽Esc-1GN(1-24)的抗氧化和抗炎活性。方法 利用MTT检测Esc-1GN(1-24)对Raw264.7和PC12细胞增殖的影响及对H_(2)O_(2)诱导PC12细胞损伤的保护作用;利用FRAP、ABTS和DPPH法检测Esc-1GN(1-24)的抗氧化活性;利用流式细胞术检测活性氧(reactive oxygen species,ROS)和细胞凋亡;通过ELISA检测Esc-1GN(1-24)对炎症因子的影响。利用小鼠足趾肿胀模型检测Esc-1GN(1-24)的体内抗炎活性。结果 Esc-1GN(1-24)细胞毒性低,并且抑制H_(2)O_(2)对PC12细胞的损伤;Esc-1GN(1-24)的总抗氧化能力强,能快速清除ABTS和DPPH自由基,减少H_(2)O_(2)诱导PC12的ROS产生和细胞凋亡;Esc-1GN(1-24)浓度依赖性抑制LPS诱导的Raw264.7细胞NO、IL-6、IL-1β和TNF-α的产生,且能抑制角叉菜胶诱导的小鼠足趾肿胀和MPO活力。结论 Esc-1GN(1-24)是一种具有良好抗氧化和抗炎活性的衍生肽,对抗菌肽的结构改造具有重要的参考意义。 Aim To investigate the antioxidant and anti-inflammatory activities of the shortened peptide Esc-1GN(1-24).Methods MTT was used to detect the effects of Esc-1GN(1-24)on Raw264.7 and PC12 cell proliferation and the protective effect on H_(2)O_(2)-induced PC12 cell injury.The antioxidant activity of Esc-1GN(1-24)was determined by FRAP,ABTS and DPPH assay.ROS and apoptosis were detected by flow cytometry.The effect of Esc-1GN(1-24)on inflammatory cytokines was detected by ELISA.Results Esc-1GN(1-24)had low cytotoxicity and could inhibit the damage of H_(2)O_(2) to PC12 cells;Esc-1GN(1-24)had a strong total antioxidant capacity,which could quickly scavenge ABTS and DPPH free radicals,and reduce H_(2)O_(2)-induced ROS production and apoptosis of PC12 cells;Esc-1GN(1-24)concentration dependably inhibited NO,IL-6,IL-1βand TNF-αproduction in LPS-induced Raw264.7 cells,and inhibited carrageenan-induced paw edema and MPO activity.Conclusions Esc-1GN(1-24)is a derivative peptide with good antioxidant and anti-inflammatory activities,which has important reference for the structural modification of antibacterial peptides.
