枸杞多糖提取工艺优化与酒泉地产枸杞中多糖及其单糖组成测定 被引量:3

Optimization of extraction process of Lycium barbarum polysaccharides and determination of polysaccharides and monosaccharides in Lycium barbarum from Jiuquan

摘要 以甘肃省酒泉地产枸杞为试验对象,采用超声辅助优化枸杞多糖提取工艺,并对酒泉地产枸杞药材中多糖及其单糖组成进行测定分析。以多糖提取率为指标,以固液比、提取温度、提取时间、超声时间为考察因素,开展单因素试验,并结合Box-Behnken响应面法优化提取工艺。按照苯酚-硫酸法测定枸杞中总多糖,利用柱前衍生化高效液相色谱(HPLC)法测定枸杞多糖的单糖组成。结果显示,枸杞多糖超声辅助最佳提取工艺为固液比1∶20(g∶mL),超声时间30 min,提取温度90℃,提取时间90 min。采用上述工艺,酒泉玉门市柳河乡产的枸杞中多糖净提取率最高,为3.83%;利用苯酚-硫酸法和柱前衍生化HPLC法分别测定9批枸杞总多糖含量和自制枸杞多糖的单糖组成,酒泉玉门市花海镇和瓜州县布隆吉乡产的枸杞中总多糖含量较高,为3.21%和3.01%;自制枸杞多糖由甘露糖、鼠李糖、半乳糖醛酸、葡萄糖、半乳糖、木糖和阿拉伯糖7种单糖组成,平均含量分别为0.62%、1.01%、3.59%、8.22%、3.59%、0.76%和4.79%,且玉门市和瓜州县产的枸杞多糖中单糖含量较高。枸杞多糖的超声辅助提取工艺稳定可靠,为其加工生产提供了理论依据,节约了时间及经济成本;对酒泉地产枸杞的多糖及单糖组成进行比较分析,可为当地枸杞特色中药材质量状况评价提供依据,有助于枸杞多糖深加工产业的发展。 In this study,Lycium barbarum from Jiuquan in Gansu Province was selected as the experimental object,the extraction process of Lycium barbarum polysaccharide(LBP)was optimized by ultrasonic-assisted extraction,and the composition of polysaccharides and monosaccharides in Lycium barbarum was determined and analyzed.Taking the extraction rate of polysaccharide as the index,solid-liquid ratio,temperature,extraction time and ultrasonic time were selected for experimental factors.The extraction process was optimized by single factor test and Box-Behnken response surface method.The polysaccharide content of Lycium barbarum was determined by phenol-sulfuric acid method,and the monosaccharide composition of LBP was determined by high performance liquid chromatography(HPLC)with pre-column derivation.The optimal ultrasonic-assisted extraction technology of LBP was as follows:the ratio of solid to liquid was 1∶20(g∶mL),the ultrasonic time was 30 min,the extraction temperature was 90℃,and the extraction time was 90 min.With the above technology,the net extraction rate of LBP from Liuhe town of Yumen City was the highest,which was 3.83%.The total polysaccharide content and monosaccharide composition of homemade LBP in 9 batches were determined by phenolsulfuric acid method and pre-column derivatization HPLC.The results showed that the total polysaccharide content in Huahai Town of Yumen City and Bulonggi Town of Guazhou County was 3.21%and 3.01%,respectively.LBP was composed of seven monosaccharides,namely mannose,rhamnose,galacturonic acid,glucose,galactose,xyloss and arabinose,with an average content of 0.62%,1.01%,3.59%,8.22%,3.59%,0.76%and 4.79%,respectively.The monosaccharides content of LBP produced in Yumen City and Guazhou County was higher.The ultrasonic-assisted extraction technology of LBP is stable and reliable,which provides theoretical basis for its processing and production,and saves time and economic cost.The analysis and comparison of polysaccharide and monosaccharide composition of Lycium barbarum in Jiuquan can provide the basis for the quality evaluation of local Lycium barbarum characteristic Chinese medicinal materials,and contribute to the development of LBP deep processing industry.
