摘要
C型凝集素是昆虫先天免疫防御系统中识别病原微生物的模式识别受体之一,具有参与细菌的识别与清除、调节血液封装和黑化等多方面的功能。本研究克隆的柞蚕凝集素基因ApCTL4-X1CDS由669个碱基构成,其编码蛋白含有223个氨基酸,具有糖识别基序QPD/WHD。通过原核表达系统表达纯化得到重组蛋白。功能实验分析显示,重组蛋白具有凝菌、抑菌、糖结合的活性,在柞蚕先天免疫防御系统中具有重要的功能。
C-type lectin is one of the pattern recognition receptors in the innate immune defense system of insects to recognize pathogenic microorganisms and has various functions such as participating in the recognition and clearance of bacteria,regulating blood encapsulation and blackening.The CDS of ApCTL4-X1 gene cloned in this study was composed of 669 bases,encoding 223 amino acids with sugar recognition motif QPD/WHD.The recombinant protein was expressed and purified by prokaryotic expression system.Functional analysis showed that the recombinant protein had the immune functions of coagulating bacteria,inhibiting bacteria and the sugar-binding activity,which played an important role in the innate immune defense system of Antheraea pernyi.
