摘要
目的:探究雅连根际土壤微生物群落结构特征及与雅连生物碱含量的相关性,揭示雅连根际土壤微生物群落结构与雅连品质的关系。方法:以二至五年生雅连及其根际土壤为材料,基于Illumina MiSeq平台,利用高通量测序技术对不同生长年限雅连根际土壤细菌16S rRNA基因和真菌ITS序列测序并进行数据分析;利用HPLC检测雅连生物碱含量。结果:各年生雅连根际土壤样本微生物的所有分类水平具有显著丰度差异;不同生长年限雅连6种生物碱总量由高到低依次为:四年生>五年生>三年生>二年生,且四年生雅连的6种生物碱总量显著高于其他各年生雅连;将雅连根际土壤中丰度前15的微生物菌属与各年生雅连生物碱指标进行Spearman分析表明,细菌标志菌属MBNT15属和分枝杆菌属Mycobacterium与雅连多种生物碱含量呈显著相关,真菌标志菌属端毛孢属Seimatosporium与雅连多个生物碱含量呈显著相关。结论:该研究揭示了雅连根际土壤微生物群落的种类与资源,发掘了与雅连生物碱含量相关的标志根际土壤微生物,为进一步研究影响雅连品质形成的核心功能微生物菌群提供了参考。
Objective:To investigate the correlation between the structural characteristics of the rhizosphere soil microbial community of Coptis deltoidea and its alkaloid content,revealing the relationship between Coptis deltoidea rhizosphere soil microbial community structure and quality.Methods:Using two to five-year-old Coptis deltoidea and its rhizosphere soil as materials,based on the Illumina MiSeq platform,high-throughput sequencing technology was used to sequence and analyze the 16S rRNA gene and fungal ITS sequence of rhizosphere soil bacteria in different growth years of Coptis deltoidea.Coptis deltoidea alkaloid content was detected by HPLC.Results:All the taxonomic levels of the microorganisms in the rhizosphere soil samples of Coptis deltoidea in different growth years had significant differences in abundance;The total content of the 6 alkaloids of Coptis deltoidea in different growth years was as follows:4 years>5 years>3 years>2 years;The total content of 6 alkaloids in four-year-old Coptis deltoidea were significantly higher than that in other years;Spearman analysis of the top 15 microbial genus in the rhizosphere soil of Coptis deltoidea and the alkaloid index of Coptis deltoidea in each year showed that the bacterial marker bacteria genus MBNT15 and Mycobacterium were significantly correlated with the contents of various alkaloids in Coptis deltoidea,and the fungal marker genus Seimatosporium was significantly correlated with the contents of several alkaloids in Coptis deltoidea.Conclusion:This study reveales the species and resources of rhizosphere soil microbial community,and excavated the marker rhizosphere soil microorganisms related to alkaloid content of Coptis deltoidea,which provides a reference for further research on the core functional microbial flora affecting the quality formation of Coptis deltoidea.
