摘要
目的目前关于鼻黏膜中Piezo1的表达研究甚少。文中旨在观察Piezo1在人鼻黏膜和鼻息肉中的表达与分布,探讨Piezo1在鼻黏膜正常生理功能中的作用。方法选取2021年8月至2022年9月南京鼓楼医院耳鼻咽喉头颈外科行鼻内手术的14例患者的下鼻甲、中鼻甲和鼻息肉组织标本。应用HE染色及免疫组化技术检测Piezo1阳性细胞的表达与分布。应用RT⁃qPCR检测Piezo1在不同部位组织间mRNA表达水平的差异及变化趋势。结果免疫组化染色结果显示,Piezo1在下鼻甲和中鼻甲中阳性细胞的表达(0.44±0.01、0.43±0.03)较鼻息肉阳性细胞的表达(0.27±0.02)显著增加(P<0.001)。RT⁃qPCR检测结果显示,Piezo1在下鼻甲、中鼻甲的mRNA表达水平(1.72±0.21、2.17±0.09)较鼻息肉(1.02±0.19)显著增加(P<0.001)。结论Piezo1在鼻黏膜广泛表达,且下鼻甲和中鼻甲Piezo1的表达较鼻息肉存在明显差异,提示Piezo1与维持人鼻黏膜正常生理功能密切相关。
Objective Little has been studied about Piezo1 expression in the nasal mucosa.The purpose of this study was to observe the expression and distribution of Piezo1 in human nasal mucosa and nasal polyp tissues,and to explore the role of Piezo1 in the normal physiological function of the nasal mucosa.Methods Tissue specimens of inferior turbinate,middle turbinate and nasal polyp tissues were selected from 14 patients who underwent intranasal surgery in the Department of Otolaryngology⁃Head and Neck Surgery of Nanjing Drum Tower Hospital from August 2021 to September 2022.The expression and distribution of Piezo1 positive cells were de⁃tected by HE staining and immunohistochemistry.The mRNA expres⁃sion levels of Piezo1 in different tissues were detected by RT qPCR.Results Immunohistochemical staining showed that the expression of Piezo1 in positive cells in inferior turbinate and middle turbinate(0.44±0.01,0.43±0.03)was significantly increased compared with that in positive cells in nasal polyp tissues(0.27±0.02)(P<0.001).RT qPCR results showed that the mRNA expression level of Piezo1 in inferior turbinate and middle turbinate(1.72±0.21,2.17±0.09)was significantly increased compared with that in nasal polyp tissues(1.02±0.19)(P<0.001).Conclusion Piezo1 is widely expressed in nasal mucosa,and the expression of Piezo1 in inferior turbi⁃nate and middle turbinate is significantly different from that in nasal polyp tissues,suggesting that Piezo1 is closely related to the main⁃tenance of normal physiological function of human nasal mucosa.
