摘要
美洲莲仅分布于北美洲和中美洲地区,其独特的淡黄色花色显著区别于亚洲莲的花色,因此在国内花卉市场显得尤其珍贵,并可作为良好的育种亲本。但由于美洲莲的不同居群在花色深浅和花被形态方面存在一定的多样性,并且随着亚美杂交莲品种越来越多,部分品种在形态上同美洲莲非常近似,可能导致资源圃中的美洲莲亲本出现不纯的情况。因此,如何鉴定纯种的美洲莲一直是困扰荷花育种和花卉企业的一个重要问题。本研究发现美洲莲APETALA 3(AP3)基因组ATG下游1107~1192 bp处存在86 bp碱基缺失,利用此特点。建立了一种快速鉴定美洲莲基因型的分子检测方法,在种子苗期或种藕阶段即可快速进行美洲莲纯种的鉴定,该方法的建立将在莲属资源的科学保护及管理、保证取材的准确性、育种和资源交流过程中具有很好的应用潜力。
American lotus(Nelumbo lutea)is only distributed in North and Central America,and its unique yellowish flower color is significantly different from that of Asian lotus,so it is especially precious in flower market and can be used as an excellent breeding parent.However,due to the diversity of flower color and morphology among different populations of American lotus,and more and more Asian-American hybrid lotus cultivars,some of which are very similar to American lotus in morphology,resulting in impurity of American lotus in germplasm resources.How to identify pure American lotus has always been an important problem for lotus breeders.A deletion of 86 bp at 1107~1192 bp position downstream from ATG codon of APETALA 3(AP3)genome was discovered and based on this,we established a molecular identification method for rapid identification of American lotus genotypes at seedling or rhizome stage.This method will have potential application in scientific protection for American lotus resources,and will be of great value in lotus breeding and resource exchange in the future.
