β-catenin、Biglycan蛋白在结肠癌中的表达及临床意义

摘要 目的观察β-链蛋白(β-catenin)、双糖链蛋白聚糖(Biglycan)蛋白在结肠癌中的表达特点及临床意义,探讨两项指标与结肠癌临床表现、病理特征的关系及β-catenin阳性表达率与微卫星状态的关系。方法选取2019年1月至2020年6月孝感市中心医院收治的80例结肠癌患者作为研究对象,收集其根治术后结肠癌标本及其癌旁正常肠组织,采用免疫组织化学染色方法检测β-catenin、Biglycan蛋白,以癌旁正常肠组织为对照,光镜下评估各项指标表达情况。采用PCR方法检测分析微卫星状态并分组。结果与癌旁正常组织相比,Biglycan蛋白在结肠癌组织中的阳性表达率明显升高,Biglycan蛋白表达与结肠癌组织的组织学分级、浸润深度、淋巴结转移及TNM分期有关,表达率差异有统计学意义(P<0.05)。与癌旁正常组织相比,β-catenin蛋白在结肠癌组织中的阳性表达率明显升高,β-catenin蛋白阳性表达与癌组织的浸润深度、淋巴结转移及TNM分期有关,表达率差异有统计学意义(P<0.05)。β-catenin阳性表达率升高与Biglycan蛋白阳性表达存在一定的相关性(r=0.922)。β-catenin蛋白阳性表达在结肠癌中MSI-H与MSI-L/MSS两组间无明显相关性,差异无统计学意义(P>0.05)。结论Biglycan与β-catenin在结肠癌组织中阳性表达率明显高于癌旁正常组织,二者表达与癌组织的浸润深度、淋巴结转移及TNM分期有关,可作为评估结肠癌预后指标,用于指导临床治疗,且二者表达相互间存在一定相关性,可能参与结肠癌的WNT信号通路激活。 【Objective】To observe the expression and clinical significance ofβ-catenin and Biglycan protein in colon cancer,and to explore the relationship between the two indicators and clinical manifestations and pathological features of colon cancer,and the relationship between the positive expression rate ofβ-catenin and microsatellite status.【Methods】In this study,80 cases of colon cancer patients admitted to our hospital were selected as the research objects.Colon cancer specimens and adjacent normal intestinal tissues after radical resection were collected.β-catenin and Biglycan proteins were detected by immunohistochemical staining.The status of microsatellites was detected and analyzed by PCR and grouped.【Results】The expression rate of Biglycan protein in colon cancer tissues was significantly higher than that in normal adjacent tissues.Biglycan protein expression was related to histological grade,invasion depth,lymph node metastasis and TNM stage of cancer tissues,and the difference in expression rate was statistically significant(P<0.05).The positive expression rate ofβ-catenin protein in colon cancer tissues was significantly higher than that in adjacent normal tissues.The positive expression rate ofβ-catenin protein was related to the depth of invasion,lymph node metastasis and TNM stage of cancer tissues,and the difference of expression rate was statistically significant(P<0.05).There was a correlation between the expression rate ofβ-catenin and the positive expression of Biglycan protein(r=0.922).β-catenin protein positive expression in colon cancer was not significantly correlated between MSI-H and MSI-L/MSS groups,and the difference was not statistically significant(P>0.05).【Conclusion】The positive expression rate of Biglycan andβ-catenin in colon cancer tissues is significantly higher than that in adjacent normal tissues.The expression of Biglycan andβ-catenin is related to the depth of invasion,lymph node metastasis and TNM stage of the cancer tissues,which may be used to evaluate the prognosis of colon cancer and guide clinical treatment,and there is a certain correlation between the expression of Biglycan andβ-catenin.May be involved in the activation of WNT signaling pathway in colon cancer.
