甲状腺切除术对甲状腺癌荷瘤小鼠的免疫应答反应的影响

摘要 目的研究甲状腺切除术对甲状腺癌荷瘤小鼠的免疫应答反应的影响。方法60只小鼠,采用随机数字表法分为对照组、模型组和手术组,每组大鼠20例。模型组采用肿瘤细胞注射进行甲状腺癌模型制备,手术组对模型组小鼠进行甲状腺切除术,比较3组小鼠的sFas、sFasL、VEGF水平、炎性因子水平、T细胞以及B细胞的增殖活性、免疫细胞水平之间的差异。结果3组小鼠sFas(F=11.231,P=0.000)、sFasL(F=6.672,P=0.000)、VEGF(F=12.231,P=0.000)之间的差异存在统计学意义,通过两两比较,3组小鼠的sFas、sFasL、VEGF水平从高到低依次为模型组、手术组以及对照组;3组小鼠的TNF-α(F=11.549,P=0.000)、IL-1β(F=12.598,P=0.000)、IL-6(F=8.997,P=0.000)之间的差异存在统计学意义,通过两两比较,3组小鼠的炎性因子水平从高到低依次为模型组、手术组以及对照组;3组小鼠的SI(F=7.006,P=0.000)、T细胞增殖活性(F=6.111,P=0.000)、B细胞增殖活性(F=8.098,P=0.000)之间的差异存在统计学意义,经过两两比较,3组小鼠的SI、T细胞增殖活性、B细胞增殖活性从高到低依次为对照组、手术组以及模型组;3组小鼠的CD3^(+)T细胞、CD8^(+)T细胞、Treg、NK之间的差异不存在统计学意义(P>0.05),3组小鼠的CD4^(+)T细胞(F=12.090,P=0.000)、CD4^(+)/CD8^(+)(F=12.443,P=0.000)、MDSC(F=12.112,P=0.000)、B细胞水平(F=12.145,P=0.000)之间的差异存在统计学意义,通过两两比较,CD4^(+)T细胞、CD4^(+)/CD8^(+)、B细胞水平高到低依次为对照组、手术组以及模型组,MDSC从高到低依次为模型组、手术组以及对照组。结论甲状腺癌荷瘤小鼠进行甲状腺手术全切后,可能通过上调CD4^(+)T细胞、CD4^(+)/CD8^(+)、B细胞水平,下调MDSC水平,进而抑制肿瘤细胞的免疫逃逸作用。 Objective To study the effect of thyroidectomy on the immune response of thyroid cancer bearing mice.Methods 60 mice were randomly divided into the control group,model group and the operation group,with 20 rats in each group.The model group was prepared with tumor cell injection,while the operation group was treated with thyroidectomy.The levels of sFas,sFasL,VEGF,inflammatory factors,the proliferative activity of T cells and B cells and the level of immune cells were compared among the 3 groups.Results The differences of sFas(F=11.231,P=0.000),sFasL(F=6.672,P=0.000)and VEGF(F=12.231,P=0.000)were statistically significant.By pairwise comparison,the levels of sFas,sFasL and VEGF in the 3 groups from high to low were model group,the operation group and the control group;the levels of TNF-α(F=11.549,P=0.000),IL-1β(F=12.598,P=0.000),IL-6(F=8.997,P=0.000,P=0.000)were higher than those of control group,Through pairwise comparison,the levels of inflammatory factors in the 3 groups of the experimental mice from high to low were model group,the operation group and the control group;Si(F=7.006,P=0.000),T cell proliferation activity(F=6.111,P=0.000)and B cell proliferation activity(F=8.098,P=0.000)were significantly different from each other,After pairwise comparison,the Si,T cell proliferation activity and B cell proliferation activity of the 3 groups of the experimental mice were in the order of the control group,the operation group and the model group from high to low;there was no significant difference in CD3^(+)T cells,CD8^(+)T cells,Treg and NK among the 3 groups(P>0.05),The percentage of CD4^(+)T cells(F=12.090,P=0.000),CD4^(+)/CD8^(+)(F=12.443,P=0.000),MDSC(F=12.112,P=0.000)and B cell ratio(F=12.145,P=0.000)were detected,By pairwise comparison,the proportion of CD4^(+)T cells,CD4^(+)/CD8^(+),B cells from high to low was control group,operation group and model group,and MDSC from high to low was the model group,the operation group and the control group.Conclusion After total thyroidectomy,the proportion of CD4^(+)T cells,CD4^(+)/CD8^(+),B cells and the proportion of MDSC may be increased and the immune escape effect of tumor cells can be inhibited.
