泛素E3连接酶膜相关锌指蛋白2高表达与胃癌临床分期、侵袭性及预后的相关性

Correlation of High Expression of Membrane-associated RING CH 2 with Clinical Stage,Tumor Invasion and Prognosis of Gastric Cancer Patients

目的研究泛素E3连接酶膜相关锌指蛋白2(MARCH2)高表达与胃癌分期、侵袭及预后之间的关系。方法将96例行胃癌根治术的胃癌患者纳为研究对象,免疫组织化学法检测患者胃癌组织及癌旁组织内MARCH2表达情况,分析MARCH2阳性表达与患者临床病理特征之间的关系。术后随访5~47个月,采用Kaplan-Meier法分析胃癌组织内MARCH2表达情况与患者生存时间的关系。结果胃癌组织内MARCH2阳性表达率高于癌旁组织,差异具有统计学意义(P<0.05)。胃癌组织内MARCH2阳性表达与胃癌浸润深度以及淋巴结转移有关(P<0.05),与患者年龄、性别、肿瘤大小、TNM分期以及分化程度无关(P>0.05)。将胃癌组织内MARCH2中、强阳性表达者纳为MARCH2强阳性表达组,其余患者纳为对照组,随访5~47个月,平均(34.51±6.16)个月,绘制生存曲线发现,强阳性组患者生存时间短于对照组,差异具有统计学意义(Rank=8.923,P=0.003)。结论胃癌组织内MARCH2阳性表达率明显上升,且MARCH2阳性表达与肿瘤浸润深度及淋巴结转移间存在密切联系,MARCH2高表达者预后生存时间更短,提示MARCH2可能参与胃癌的发生及发展。

Objective To analyze the correlation of high expression of membrane-associated RING-CH 2(MARCH2)with clinical stage,tumor invasion and poor prognosis of gastric cancer patients.Methods A sample of 96 patients with gastric cancer receiving radical gastrectomy were enrolled.The expression of MARCH2 in gastric cancer tissues and adjacent normal tissues was detected using immunohistochemistry,and the relationship between the positive expression of MARCH2 and clinicopathological characteristics of patients was discussed.Patients were followed up for 5~47 months after operation,and the Kaplan-Meier method was used to analyze the relationship between MARCH2 expression and survival time of gastric cancer patients.Results The positive expression rate of MARCH2 in gastric cancer tissues was higher than that in adjacent normal tissues(P<0.05).The positive expression of MARCH2 in gastric cancer tissues was correlated with the tumor invasion depth and lymph node metastasis(P<0.05),while had no correlation with the age,gender,tumor size,TNM stage and differentiation degree of patients(P>0.05).Patients with moderate-to-strong positive expression of MARCH2 in gastric cancer tissues were included as strong positive group,and the other patients were included as the control group.The follow-up time ranged from 5 to 47 months,with a mean of(34.51±6.16)months.Survival curves revealed that patients in strong positive group had shorter survival times than that in control group,with a statistical difference(Rank=8.923,P=0.003).Conclusion MARCH2 positive expression rate is notably increased in gastric cancer tissues,and is closely related to the tumor invasion depth and lymph node metastasis.Moreover,patients with high expression of MARCH2 have short survival time,suggesting that MARCH2 may be involved in the occurrence and development of gastric cancer.
