不同麻醉预处理对肾脏缺血再灌注损伤模型的肾脏保护作用及分子机制研究

摘要 目的分析不同麻醉药物处理后对肾脏缺血再灌注损伤的保护作用及分子机制。方法于2019年1—6月吉林省通化市人民医院,将60只SD大鼠分为NC组、Sham组、I/R组、Pro组、Rem组和Sev组,每组10只,检测各组BUN、肌酐(Cr)、白细胞介素-1β(IL-1β)、白细胞介素-8(IL-8)、超氧化物歧化酶(SOD)含量、丙二醛(MDA)水平、细胞凋亡情况、肾组织自噬相关蛋白的表达。结果I/R组BUN、Cr、IL-1β、IL-8、SOD、MDA,与NC组、Sham组比较,差异有统计学意义(P<0.05)。Pro组、Rem组及Sev组的BUN、Cr、IL-1β、SOD与I/R组比较,差异有统计学意义(P<0.05)。Sev组BUN、Cr、MDA,与Pro组、Rem组比较,差异有统计学意义(P<0.05)。I/R组细胞凋亡明显,Rem组凋亡指数(AI)(0.445±0.072)低于I/R组(0.714±0.063),差异有统计学意义(P<0.05)。Sev组AI水平(0.311±0.078)低于Pro组的(0.496±0.081)、Rem组的(0.445±0.072),差异有统计学意义(P<0.05)。I/R组及Sev组自噬小体数量升高,I/R组pAMPK、pmTOR及Caspase-3均高于NC组、Sham组、Pro组、Rem组及Sev组,差异有统计学意义(P<0.05)。结论3种不同麻醉药物包括丙泊酚、瑞芬太尼及七氟醚,在缓解肾脏缺血再灌注损伤方面均具有一定的作用。 Objective To analyze the protective effect and molecular mechanism of different anesthetic drugs on renal ischemia-reperfusion injury.Methods At the People´s Hospital of Tonghua City,Jilin Province from January to June 2019,60 SD rats were divided into NC group,Sham group,I/R group,Pro group,Rem group,and Sev group,with 10 rats in each group.BUN,creatinine(Cr),and interleukin-1 were detected in each groupβ(IL-1β)、Interleukin-8(IL-8),superoxide dismutase(SOD)content,malondialdehyde(MDA)level,apoptosis,expression of autophagy related proteins in renal tissue.Results BUN,Cr,IL-1β,IL-8,SOD and MDA in I/R group were significantly different from those in NC group and Sham group,and the difference was statistically significant(P<0.05).There were significant dif-ferences in BUN,Cr,IL-1βand SOD between Pro group,Rem group and Sev group and I/R group,and the difference was statistically significant(P<0.05).BUN,Cr and MDA in the Sev group were significantly different from those in the Pro group and the Rem group,and the difference was statistically significant(P<0.05).The apoptosis index(AI)of Rem group(0.445±0.072)was lower than that of I/R group(0.714±0.063),and the difference was statistically signifi-cant(P<0.05).The AI level of Sev group(0.311±0.078)was lower than that of Pro group(0.496±0.081)and Rem group(0.445±0.072),and the difference was statistically significant(P<0.05).The number of autophagosomes in I/R group and Sev group increased.The pAMPK,pmTOR and Caspase-3 in I/R group were higher than those in NC group,Sham group,Pro group,Rem group and Sev group,and the difference was statistically significant(P<0.05).Conclusion Three different anesthetic drugs,including propofol,remifentanil and sevoflurane,all have a certain effect on alleviating renal ischemia-reperfusion injury.
