腹腔注射鸡卵白蛋白对小鼠呼吸道致敏模型的影响 被引量:1

Effect of intraperitoneal injection of ovalbumin on mouse respiratory sensitization model

摘要 目的探讨腹腔注射鸡卵白蛋白(ovalbumin,OVA)及OVA联合免疫佐剂对小鼠呼吸道接触OVA引起的致敏作用的影响。方法将BALB/c小鼠分为空白对照组、阴性对照组(腹腔注射生理盐水)、OVA组(腹腔注射OVA)、OVA^(+)Al(OH)_(3)组[腹腔注射OVA和Al(OH)_(3)],每组8只。腹腔注射后第12天,除空白对照组,其余3组小鼠气管滴下OVA,连续3 d,第15天用流式细胞仪测定肺泡灌洗液(bronchoalveolar lavage,BAL)和肺纵膈淋巴结(mediastinal lymph node,MLN)中的免疫细胞(CD45^(+)细胞)数目,中性粒细胞(Gr1^(+)细胞)、T细胞(CD3^(+)细胞)、B细胞(CD19^(+)细胞)以及CD4^(+)T细胞中调节性T细胞(CD25^(+)Foxp3^(+)细胞)的百分比。结果与空白对照组相比,气管滴下OVA后,阴性对照组、OVA组和OVA^(+)Al(OH)_(3)组BAL中CD45^(+)细胞数目、CD45^(+)细胞中中性粒细胞和T细胞的百分比以及数目显著增高(P<0.05);同时MLN的T和B细胞数目显著增多,T细胞百分比显著降低而B细胞百分比显著升高,CD4^(+)T细胞中调节性T细胞百分比显著降低(P<0.05)。与阴性对照组相比,腹腔注射OVA和OVA联合Al(OH)_(3)降低了MLN中T细胞百分比而升高了B细胞百分比(P<0.05)。与单独腹腔注射OVA相比,腹腔注射OVA联合Al(OH)_(3)增加了MLN的T和B细胞的数目(P<0.05)。结论在呼吸道接触OVA引起的小鼠致敏模型中,腹腔注射OVA没有影响呼吸道的免疫细胞构成比和数目,但影响了肺纵膈淋巴结的T和B细胞的百分比,而OVA联合免疫佐剂比单独注射OVA更增加了T和B细胞数目。 Objective This study aims to investigate the effects of intraperitoneal injection of ovalbumin(OVA)alone or in combination with an immune adjuvant on sensitization caused by respiratory exposure to OVA in mice.Methods BALB/c mice were divided into the following groups:blank control group,negative control group(intraperitoneal injection of normal saline),OVA group(intraperitoneal injection of OVA),and OVA^(+)Al(OH)_(3)group[intraperitoneal injection of OVA and Al(OH)_(3)],with eight mice in each group.On the 12th day after intraperitoneal injection,except for the blank control group,mice in the other three groups were exposed to OVA via tracheal drops for 3 consecutive days.On the 15th day,flow cytometry was used to measure the number of immune cells(CD45^(+)cells)in bronchoalveolar lavage(BAL)and mediastinal lymph nodes(MLN).The percentages of neutrophils(Gr1^(+)cells),T cells(CD3^(+)cells),B cells(CD19^(+)cells),and regulatory T cells(CD25^(+)Foxp3^(+)cells)in CD4^(+)T cells were also determined.Results Compared with the blank control group,the negative control group,OVA group,and OVA^(+)Al(OH)_(3)group showed a significant increase in the number of CD45^(+)cells in BAL,as well as in the percentage and number of neutrophils and T cells within CD45^(+)cells after tracheal exposure to OVA(P<0.05).Moreover,the MLN exhibited a significant increase in the number of T and B cells,a significant decrease in the percentage of T cells,a significant increase in the percentage of B cells,and a significant decrease in the percentage of regulatory T cells within CD4^(+)T cells(P<0.05).Compared with the negative control group,intraperitoneal injection of OVA and OVA^(+)Al(OH)_(3)resulted in a decrease in the percentage of T cells and an increase in the percentage of B cells in MLN(P<0.05).Furthermore,intraperitoneal injection of OVA combined with Al(OH)_(3)increased the number of T and B cells in MLN compared to intraperitoneal injection of OVA alone(P<0.05).Conclusion In the mouse model of respiratory sensitization induced by OVA exposure,intraperitoneal injection of OVA does not affect the composition and number of immune cells in the respiratory tract but influences the percentages of T and B cells in the mediastinal lymph nodes.Moreover,OVA combined with an immune adjuvant increase the number of T and B cells compared to OVA alone.
