氯化胆碱基低共熔溶剂优化草珊瑚总黄酮提取工艺 被引量:1

Optimization and extraction of total flavonoids from Sarcandra glabra(Thunb.)Nakai by choline chloride-based deep eutectic solvents

摘要 为优化草珊瑚总黄酮提取工艺,采用单因素试验探究了5种氯化胆碱基低共熔溶剂、氢键受体与氢键供体的摩尔比、提取时间、液料比、提取温度对草珊瑚总黄酮得率的影响,通过L9(34)正交试验优化提取工艺。结果表明:最优工艺为氯化胆碱/乙二醇摩尔比1:25,时间60 min,液料比30:1(mL/g),温度80℃,平均得率为17.20%,RSD为0.64%。低共熔溶剂较传统溶剂(水、醇)对草珊瑚总黄酮的提取率高,氯化胆碱/乙二醇体系低共熔溶剂是一种绿色高效的提取草珊瑚总黄酮的溶剂。 Deep eutectic solvents(DES)was used to evaluate and optimize the extraction process of the total flavonoid in Sarcandra glabra(Thunb.)Nakai.First of all,5 kinds of choline chloride-based DESs were selected as potential extraction solvents,and the DES extraction effect of choline chloride/ethylene glycol was determined to be the best.Then,single-factor experiments were conducted to explore the effects of DES composition,molar ratio of hydrogen bond acceptor(HBA)to hydrogen bond donor(HBD),extraction time,and ratio of liquid to material,extraction temperature on the yield of the total flavonoid in Sarcandra glabra(Thunb.)Nakai.Based on the above experiments,the orthogonal experimental design of four factors and three levels was established.The results showed that the best extraction conditions of total flavonoids in Sarcandra glabra(Thunb.)Nakai were concluded:molar ratio of choline chloride to ethylene glycol was 1:25,extraction time was 60 min,ratio of liquid to material was 30:1(mL/g),extraction temperature was 80℃.Under these conditions,the yield of total flavonoids in Sarcandra glabra(Thunb.)Nakai was 17.20%,RSD was 0.64%.Compared with the traditional solvents(water and alcohol),the deep eutectic solvent of choline chloride/ethylene glycol system was a kind of green and efficient solvent to extract the total flavonoids of Sarcandra glabra(Thunb.)Nakai.
