摘要
蒲公英中获得强抗菌活性的内生真菌,分析其次级代谢产物的化学结构,为新颖抗菌活性天然产物的发现奠定基础。通过滤纸片法筛选获得抗菌活性目标菌株,采用形态学和分子生物学方法鉴定真菌的种属,并采用质谱-分子网络技术预测真菌的次级代谢产物的化学结构。从21株形态各异的蒲公英内生真菌中筛选获得了1株抗菌活性较好的菌株2-7,粗提物对4种金黄色葡萄球菌在10μg/片浓度下抑菌圈为10.82~17.58 mm,其种属鉴定为Alternaria sp.,该真菌可代谢产生黄酮、甾体、生物碱、脂肪酸、丁内酯、萜类等化合物,其中黄酮分子簇中的部分母离子可能为潜在的新化合物。
In this paper,endophytic fungi with strong antibacterial activity were obtained from dandelion,and the chemical structure of its secondary metabolites was analyzed,which laid the foundation for the discovery of novel natural products with antibacterial activity.The target strain with antibacterial activity was screened by filter paper method,and its species identification was carried out by morphological and molecular biological methods.The chemical structures of secondary metabolites of target fungus were predicted by mass spectrometry-molecular network technology.The strain of 2-7 was screened from 21 strains of dandelion endophytic fungi with different morphology and identified as Alternaria sp..Its inhibition zone of crude extract against four kinds of Staphylococcus aureus was 10.82~17.58 mm at the concentration of 10μg/piece.The fungus could produce flavonoids,steroids,alkaloids,fatty acids,butyrolactone,terpenoids and other compounds.Some parent ions in flavone clusters of molecular network are potential new compounds.
