缺氧诱导因子-1α调控支气管哮喘中性粒细胞凋亡的相关性研究

A correlation study of hypoxia inducible factor-1αin regulating neutrophil apoptosis in bronchial asthma

目的探讨缺氧诱导因子-1α(HIF-1α)调控支气管哮喘患者外周血中性粒细胞凋亡的相关性,为支气管哮喘的亚临床和临床治疗提供理论基础。方法选取2021年1—5月于本院诊治的30例支气管哮喘患者作为哮喘组,另选取同期于本院实施健康体检者30名作为健康组,分离纯化中性粒细胞,检测两组中HIF-1αmRNA的表达水平,流式细胞术检测中性粒细胞凋亡率。将人原髓细胞白血病细胞(HL-60细胞)诱导为类中性粒细胞,分为缺氧组和正常组,检测两组HIF-1αmRNA表达水平及类中性粒细胞凋亡率。结果哮喘组外周血中性粒细胞HIF-1α的表达水平明显高于健康组,且中性粒细胞凋亡率低于健康组,差异有统计学意义(P<0.05)。缺氧组HIF-1α表达水平高于正常组,且类中性粒细胞凋亡率低于正常组,差异有统计学意义(P<0.05)。结论HIF-1α参与调控支气管哮喘患者外周血中性粒细胞的凋亡,为支气管哮喘的亚临床和临床治疗提供理论基础。

Objective To investigate the correlation between hypoxia-inducible factor-1α(HIF-1α)in the regulation of peripheral blood neutrophil apoptosis in patients with bronchial asthma,and to provide a theoretical basis for the subclinical and clinical treatment of bronchial asthma.Methods A total of 30 patients with bronchial asthma who were treated in our hospital from January to May 2021 were selected as the asthma group,In addition,30 patients who underwent health examinations in our hospital at the same time were selected as healthy group,neutrophils were isolated and purified,the expression levels of HIF-1αmRNA in the two groups were detected,and the apoptosis rate of neutrophils was detected by flow cytometry.Human promyeloid leukemia cells(HL-60 cells)were induced to be neutrophils,divided into hypoxic group and normal group,and the expression level of HIF-1αmRNA and the apoptosis rate of neutrophils were measured in two groups.Results The expression level of peripheral neutrophil HIF-1αin the asthma group was significantly higher than that in the healthy group,and the apoptosis rate of neutrophils was lower than that in the healthy group,and the difference was statistically significant(P<0.05).The expression level of HIF-1αin the hypoxic group was higher than that in the normal group,and the apoptosis rate of neutrophil-like was lower than that in the normal group,and the difference was statistically significant(P<0.05).Conclusion HIF-1αis involved in regulating apoptosis of neutrophils in peripheral blood in patients with bronchial asthma,which provides a theoretical basis for the subclinical and clinical treatment of bronchial asthma.
