摘要
干扰素刺激基因15(interferon-stimulated gene15,ISG15)是最早发现的泛素样蛋白,在恶性肿瘤中显示出复杂的致癌或抗癌双重活性,然而其具体调控机制仍未阐明。近年来研究发现ISG15通过调控DNA复制分叉进程、自噬及线粒体功能、免疫微环境及靶蛋白的翻译后修饰等参与恶性肿瘤细胞的增殖、凋亡、侵袭转移、干细胞干性及耐药等,是潜在的诊断、预后标志物及治疗靶点。该文就ISG15调控恶性肿瘤发生发展作用机制的最新发现进行综述。
Interferon-stimulated gene 15(ISG15),the first ubiquitin-like protein identified,shows complex dual oncogenic or anticancer activities in malignant tumors.However,its specific regula tory mechanisms remain elusive.Recent studies have re-vealed that ISG15 is a potential diagnostic and prognostic marker and therapeutic target by reg ulating DNA replica tion process,autophagy and mitochondrial function,immune microenvironment and post-translational modifications of target proteins,w hich ure involved in proliferation,apoptosis,invasion and metastasis,stem cell stemness and drug resistance of malignant tumor cells.In this paper,we review the recent findings on the mechanism of ISG15's role in regulating malignant tumorigenesis and deve lopment.
