摘要
目的探讨癌症患者外周血中调节性T细胞(regulatory T cells,Tregs)亚型即1型辅助性T细胞(type 1 helper T cell,Th1)样Treg细胞(T-bet^(+)Treg)、Th2样Treg细胞(GATA3^(+)Treg)、Th9样Treg细胞(Pu-1^(+)Treg)、Th17样Treg细胞(RORγ^(+)Treg)及滤泡型辅助性T细胞(follicular helper T cell,Tfh)样Treg细胞(CXCR5^(+)Treg)的表达情况及其对应的CD4^(+)T细胞子集:Th1、2型辅助性T细胞(type 2 helper T cell,Th2)、9型辅助性T细胞(type 9 helper T cell,Th9)、17型辅助性T细胞(type 17 helper T cell,Th17)、Tfh的表达。方法将23例癌症患者设为研究组,23例门诊健康体检者设为正常对照组,采用流式细胞术检测患者外周血中T-bet^(+)Treg、GATA3^(+)Treg、Pu-1^(+)Treg、RORγ^(+)Treg、CXCR5^(+)Treg数量以及CD4^(+)T细胞子集Th1、Th2、Th9、Th17、Tfh的数量变化情况,并将二者进行对比分析。结果与对照组比较,研究组患者外周血中Treg细胞的数量显著升高,其亚型T-bet^(+)Treg、GATA3^(+)Treg、RORγ^(+)Treg的数量明显增加(P<0.05),而Pu-1^(+)Treg、CXCR5^(+)Treg数量差异无统计学意义(P>0.05)。与对照组比较,研究组外周血体内的Th1细胞减少,Th2、Th9、Th17、Tfh数量增多(P<0.05)。结论患者的抗肿瘤免疫功能可能被数量增加的Treg细胞所抑制,主要增加的Treg细胞亚型为T-bet^(+)Treg、GATA3^(+)Treg、RORγ^(+)Treg,其中T-bet^(+)Treg细胞增加最为明显。
Objective To explore the subtypes of regulatory T cells(Tregs) in the peripheral blood of cancer patients, namely the expression of type 1 helper T cell(Th1) like Treg cells(T-bet^(+)Treg), Th2 like Treg cells(GATA3^(+)Treg), Th9 like Treg cells(Pu-1^(+)Treg), Th17 like Treg cells(RORγ^(+)Treg) and follicular helper T cell(Tfh) like Treg cells(CXCR5^(+)Treg) and their corresponding CD4^(+)T cell subsets: the expression of Th1, type 2 helper T cell(Th2), type 9 helper T cell(Th9), type 17 helper T cell(Th17) and Tfh. Methods A total of 23 cancer patients were selected as the study group, and 23 healthy patients in the outpatient department were selected as the normal control group, the number of T-bet^(+)Treg, GATA3^(+)Treg, Pu-1^(+)Treg, RORγ^(+)Treg, CXCR5^(+)Treg in peripheral and the changes in number of CD4^(+) T cell subsets Th1, Th2, Th9, Th17 and Tfh were detected by flow cytometry, and the two were compared and analyzed. Results Compared with the control group, the number of Treg cells in peripheral blood of patients in study group significantly increased, and the number of T-bet^(+)Treg, GATA3^(+)Treg、RORγ^(+)Treg significantly increased(P0.05), while there was no significant difference in the number of Pu-1^(+)Treg and CXCR5^(+)Treg(P0.05). Compared with the control group, the number of Th1 cells in the peripheral blood of the study group decreased, while the number of Th2,Th9,Th17 and Tfh increased(P0.05). Conclusion The anti-tumor immune function of cancer patients may be inhibited by the increased number of Treg cells. The main increased Treg cell subtypes are T-bet^(+)Treg, GATA3^(+)Treg and RORγ^(+)Treg, among which the increase of T-bet^(+)Treg cells is the most obvious.
