摘要
重组酶介导的扩增(recombinase-aid amplification,RAA)是一种新型等温体外核酸扩增方法,操作简单、反应效率高、灵敏度高。由于细菌死亡后其DNA在环境中可存在较长时间,利用RAA检测目标DNA片段极易产生假阳性结果,造成不必要的损失。因此,建立基于DNA扩增策略的活菌检测方法对食品加工、环境监测等多个领域具有重要意义。本文综述了RAA技术的原理及在病原微生物检测方面的应用,以及活菌检测技术的研究进展,以期为开发基于RAA技术的活菌检测方法提供理论指导。
Recombinase-aid amplification(RAA) is a novel isothermal nucleic acid amplification method in vitro. The detection method based on RAA technology has the advantages of simple operation,high reaction efficiency and high sensitivity. Since the DNA of bacteria remains in the environment for a long time after death,the detection of target DNA fragments using RAA technology can easily produce false positive results and cause unnecessary losses.Therefore,the establishment of a viable bacteria detection method based on DNA amplification strategy is of great significance to the food processing industry,environment monitoring and other areas. This paper reviewed the principle and application in detection of pathogenic microorganisms of RAA technology,as well as the research progress of viable bacteria detection technology,in order to provide theoretical guidance for the development of viable bacteria detection methods based on RAA technology.
