盐酸小檗碱脂质体的质量评价和抗H_(22)肿瘤药效学初步探讨 被引量:1

摘要 目的采用改良pH梯度法制备盐酸小檗碱脂质体,并对其质量和抗H_(22)肿瘤效果进行初步评价。方法通过单次注射盐酸小檗碱脂质体考察其对健康KM小鼠的急性毒性,并连续多次静脉注射研究其对荷H_(22)肿瘤KM小鼠的治疗效果。结果盐酸小檗碱溶液的LD_(50)值为7.24 mg/kg,而盐酸小檗碱脂质体给药剂量为35 mg/kg时仍无小鼠死亡现象发生。以10 mg/kg和20 mg/kg剂量给药时,盐酸小檗碱脂质体的质量抑瘤率分别为40.72%和51.57%。结论盐酸小檗碱脂质体的毒性较低,具有显著的抑制H 22肿瘤增长作用。 Objective To prepare berberine hydrochloride liposomes by modified pH gradient method and preliminarily evaluate its quality and anti-H 22 tumor effect.Methods The acute toxicity of berberine hydrochloride liposome to healthy KM mice was investigated by single injection,and the therapeutic effect of berberine hydrochloride liposome on KM mice with H_(22) tumor was studied by continuous intravenous injection.Results The LD_(50) of berberine hydrochloride solution was 7.24 mg/kg,but no death occurred when the dosage of berberine hydrochloride liposome was 35 mg/kg.The mass inhibitory rates of berberine hydrochloride liposome were 40.72% and 51.57%when administered at 10 mg/kg and 20 mg/kg,respectively.Conclusion Berberine hydrochloride liposome has low toxicity and significant inhibition of H_(22) tumor growth.
