牛源铜绿假单胞菌的分离鉴定及中草药抑杀效果比较分析

摘要 为查明养殖场引起牛只呼吸道疾病的病原菌,采集患病牛鼻拭子,采用实验室方法进行细菌的分离鉴定;通过细菌毒力基因的检测、致病性试验、药敏试验分析分离菌生物学特性;并明确22种中草药对其抑杀效果。结果表明,分离菌为两端钝圆革兰氏阴性短杆菌,16SrRNA基因序列与GeneBank铜绿假单胞菌比对相似序列达到100%,表明分离菌株为铜绿假单胞菌;菌株携带Exo S、Exo Y毒力基因,8h内可致小鼠死亡。药敏试验结果显示,该菌对环丙沙星极为敏感,对阿米卡星、新霉素、复方新诺明等9种药物中度敏感;中草药药敏试验结果显示,乌梅、五倍子、五味子对分离菌具有较好的抑菌作用,其MIC分别为31.25mg/mL和62.50mg/mL,对病原菌的生长繁殖具有较好抑制作用。本研究为该病的防控提供参考。 To find out the pathogenic bacteria causing respiratory diseases of cattle in the breeding farm,the nose swabs of sick cattle were collected,and the bacteria were isolated and identified by laboratory methods.Detection of bacterial virulence gene,pathogenicity test and drug sensitivity test were carried out to analyze the biological characteristics of the isolated bacteria.The inhibition and killing effects of 22 kinds of Chinese herbal medicines were also determined.The results showed that the isolated bacteria were Gram negative bacilli with round ends.The 16S rRNA gene sequence was 100%similar to that of Pseudomonas aeruginosa in GeneBank.The strain carried Exo S and Exo Y virulence genes,which could kill mice within 8 hours.The results of drug sensitivity test showed that it was extremely sensitive to ciprofloxacin,and moderately sensitive to 9 drugs,such as amikacin,neomycin,and compound sulfamethoxazole.The results of drug sensitivity test of traditional Chinese medicine showed that Fructus Mume and Fructus Schisandrae Chinensis had good antibacterial effect on the isolated bacteria.Its MIC and MBC are 31.25 mg/mL and 62.50 mg/mL respectively.It indicated that the isolated strain was Pseudomonas aeruginosa,and the above drugs had a good inhibitory effect on the growth and reproduction of pathogenic bacteria.This study provides a reference for the prevention and control of the disease.
