双歧杆菌乳杆菌三联活菌片联合二甲双胍对2型糖尿病小鼠糖代谢的影响 被引量:2

摘要 目的探讨双歧杆菌乳杆菌三联活菌片联合二甲双胍对2型糖尿病(T2DM)小鼠糖代谢的影响。方法选取昆明小鼠80只,给予高脂饲料+10%蔗糖溶液喂养2周,采用四氧嘧啶200 mg/kg腹腔注射建立糖尿病小鼠模型,并将造模后小鼠的饮水更换为25%蔗糖溶液。在四氧嘧啶腹腔注射后的第7天,测定小鼠餐后2 h血糖,以餐后2 h血糖水平≥7.8 mmol/L视为模型建立成功。造模成功后的小鼠分别给予0.9%氯化钠溶液(模型组),二甲双胍(400 mg/kg)(二甲双胍组),二甲双胍联合双歧杆菌乳杆菌三联活菌片剂量低(1×10^(7) CFU/kg)(低剂量组)、中(2×10^(7) CFU/kg)(中剂量组)、高(3×10^(7) CFU/kg)(高剂量组)灌胃,连续3周后检测小鼠餐后2 h血糖,进行小鼠胰腺组织病理学检查及小鼠肠道菌群的丰富度分析。结果给药后,与模型组比较,给药组餐后2 h血糖均下降(均P<0.05);与二甲双胍组比较,高剂量组的餐后2 h血糖下降程度有统计学意义(P<0.05);高剂量组对T2DM小鼠的胰腺组织病理学评分与模型组比较有统计学意义(P<0.05);高剂量组小鼠肠道菌群Chao1指数高于低、中剂量组和二甲双胍组且具有统计学意义(P<0.05)。结论与单用二甲双胍比较,高剂量双歧杆菌乳杆菌三联活菌片(3×10^(7) CFU/kg)联用二甲双胍能增加小鼠肠道菌的丰富度,更好地保护小鼠胰腺组织,发挥调节T2DM小鼠糖代谢作用,从而控制血糖水平。 Objective To investigate the effect of bifidobacterium lactobacillus triple viable tablet combined with metformin on glucose metabolism in type 2 diabetes mice.Methods 80 Kunming mice were fed with highfat diet plus 10%sucrose solution for 2 weeks.The diabetes mice model was established by intraperitoneal injection of alloxan 200 mg/kg,and the drinking water of the mice was changed to 25%sucrose solution after modeling.On the 7th day after intraperitoneal injection of alloxan,the 2 h postprandial blood glucose levels of mice were measured,and a 2 h postprandial blood glucose level of≥7.8 mmol/L was considered as a successful model establishment.After successful modeling,mice were given 0.9%sodium chloride solution(model group),metformin(400 mg/kg,metformin group),and a low dose of metformin combined with bifidobacterium lactobacillus triple viable tablets(1×10^(7)CFU/kg)(low-dose combination therapy group),medium(2×10^(7)CFU/kg)(medium-dose group of combination medication),high(3×10^(7)CFU/kg)(high-dose group of combined drugs).After 3 consecutive weeks,the mice were tested for 2 h postprandial blood glucose,histopathological examination of the mouse pancreas and analysis of the abundance of intestinal flora in mice.Results After administration,compared with the model group,the decrease in blood glucose at 2 hours after meal in the administration group was statistically significant(P<0.05);compared with the metformin group,the high-dose group had a lower and statistically significant 2-hour postprandial blood glucose decrease(P<0.05);the histopathology examination and histopathological score analysis of the mouse pancreas showed that the combination of high-dose of drugs had the best protection on the pancreatic tissue of T2DM mice;the Chao1 index of intestinal microbiota in the high-dose group of mice was higher than that in the low and medium dose groups and the metformin group,with statistical significance(P<0.05).Conclusion Compared with metformin alone,high-dose bifidobacterium lactobacillus triple viable tablets(3×10^(7) CFU/kg)combined with metformin can increase the abundance of intestinal flora in mice,better protect mouse pancreatic tissue,and play a better role in regulating glucose metabolism in T2DM mice,thus controlling blood glucose levels.
