穿山龙总皂苷对高尿酸血症大鼠肠道菌群和短链脂肪酸代谢的影响 被引量:1

Effects of Total Saponins of Rhizoma Dioscorea Nipponica on Intestinal Flora and Short-Chain Fatty Acid Metabolism in Rats with Hyperuricemia

摘要 目的:探究穿山龙总皂苷对高尿酸血症(HUA)大鼠肠道菌群及短链脂肪酸的影响。方法:30只SD大鼠随机分为3组,分别为空白组(Control)、模型组(Model)、穿山龙总皂苷组(TSRDN),每组10只。空白组正常饲养,其余组连续35 d给予10%酵母饲料和5%氧嗪酸钾溶液进行HUA模型复制,从第15~35天,穿山龙总皂苷组按照480 mg/kg的剂量灌胃给予穿山龙总皂苷溶液。结果:连续造模14 d即可复制HUA模型,SUA、SBUN水平升高(P<0.01)。连续给药干预21 d,穿山龙总皂苷能够显著降低SUA、SBUN水平(P<0.01)。HUA大鼠肠道菌群中的厚壁菌门和拟杆菌门的丰度相对较高(P<0.01)。穿山龙总皂苷能显著升高HUA大鼠肠道菌群厚壁菌门、乳酸杆菌属、梭菌属、瘤胃菌属、鼠李糖乳杆菌种、约氏乳杆菌种、罗伊氏乳杆菌种的丰度(P<0.05,P<0.01)。利用GC-MS技术检测大鼠粪便中SCFA的含量变化,结果显示穿山龙总皂苷能显著提升HUA大鼠粪便中丙酸、丁酸的含量(P<0.01)。结论:穿山龙总皂苷可以调控菌群代谢产物SCFA的含量变化,促进有益菌在肠道内繁殖,同时抑制有害菌的生长。 Objective:To study the effects of total saponins of Rhizome Dioscorea Nipponica(TSRDN)on intestinal flora and short-chain fatty acid in rats with hyperuricemia.Methods:30 SD rats were randomly divided into control,model,and TSRDN groups.The rats in control group were fed normally,and the other groups were given 10%yeast diet and 5%potassium oxyazinate solution for 35 consecutive days to replicate HUA model.From day 15 to 35,TSRDN group was administered by gavage at a dose of 480 mg/kg of solution of TSRDN.Results:HUA model was replicated after 14 days continuous modeling,and the levels of SUA and SBUN increased(P<0.01).After the drug had been continuously administered for 21 days,the TSRDN could significantly lower the levels of SUA and SBUN.The abundance of firmicutes and bacteroidetes in the gut microbiota of HUA rats was relatively high(P<0.01).The TSRDN could significantly increase the abundance of firmicutes,lactobacillus,clostridium,ruminococcus,lactobacillus rhamnosus,lactobacillus johnsonii and lactobacillus reuteri in the intestinal flora of hyperuricemia rats(P<0.01).The changes in SCFA content of rat feces were detected through GC-MS technology,and the results showed that TSRDN could significantly improve the contents of propionic acid and butyric acid in HUA rat feces.Conclusion:TSRDN can regulate the content change of SCFA,a metabolite of intestinal flora,promote the reproduction of beneficial bacteria in the intestinal tract,and inhibit the growth of harmful bacteria.
