摘要
采用超声辅助提取法、回流提取法和冷浸提取法3种方法提取百合鳞茎中总黄酮,比较不同提取方法总黄酮得率,通过单因素实验和正交实验优化提取条件,并对不同产地、不同品种百合鳞茎总黄酮含量进行分析。结果表明3种提取方法总黄酮得率大小依次为超声辅助提取法、回流提取法和冷浸提取法。百合总黄酮最佳提取工艺条件为乙醇浓度70%,料液比1∶30(g∶mL),超声温度70℃,超声提取40 min。采用最佳提取工艺对百合鳞茎总黄酮含量进行测定,结果表明不同产地和不同品种百合鳞茎总黄酮含量存在一定差异。四个百合品种总黄酮含量依次为:卷丹百合>兰州百合>川百合>龙牙百合。不同产地百合鳞茎总黄酮含量依次为:江苏宜兴>湖北恩施>兰州百合>四川江油>云南曲靖>江西宜春。本研究为百合总黄酮提取和百合资源的综合开发利用提供了参考依据。
Three kinds of extraction methods:reflux extraction method,cold maceration method,ultrasonic extraction method were used to extract the content of total flavonoids in Lily.The extraction rate of total flavonoids from different extraction methods was compared,through single factor experiment and orthogonal experiment the extraction conditions of total flavonoids were optimized and the differences of total flavonoids from lily bulbs of different origin and varieties were analyzed.The results showed that the total flavonoids extraction rate of the three extraction methods was ultrasonic extraction method,reflux extraction method and cold maceration method.The optimum extraction conditions of total flavonoids from lily bulbs was as follows:ethanol concentration of 70%,solid-liquid ratio of 1∶30 g/mL,ultrasonic temperature of 70℃and ultrasonic extraction for 40 min.The optimal technology was used to determine the content of total flavonoids in lily bulbs.The results showed that there were some differences in the content of total flavonoids in lily bulbs from different producing areas and different varieties.The results show that L.lancifolium content of total flavonoids in four Lilium varieties was the highest,followed by L.davidii var.Unicolor and L.davidii,the L.browniivar.viridulum was lowest.The contents of total flavonoids in six lilium producing areas were successively:Jiangsu Yixing>Hubei Enshi>Gansu Lanzhou>Sichuan Jiangyou>Yunnan Qujing>Jiangxi Yichun.This study provided a reference for the extraction of total flavonoids from lily and the comprehensive development and utilization of lily resources.
