酶解法与乙醇渗漉法制备颠茄流浸膏的比较研究 被引量:1

摘要 目的比较研究酶解法与乙醇渗漉法提取颠茄草有效成分的提取效率。方法采用酶解法对颠茄草进行提取,筛选纤维素酶、淀粉酶、果胶酶和植物提取复合酶,用高效液相色谱法(HPLC)测定颠茄流浸膏中硫酸天仙子胺和东莨菪内酯的含量,以乙醇渗漉法为对照,比较2种方法的提取效果。结果筛选出植物提取复合酶为最适酶,采用植物提取复合酶的酶解法提取硫酸天仙子胺和东莨菪内酯的收率分别为95.57%和94.50%,乙醇渗漉法提取硫酸天仙子胺和东莨菪内酯的收率分别为85.43%和85.98%,酶解法提取收率优于乙醇渗漉法。结论酶解法制备颠茄流浸膏的提取效率优于乙醇渗漉法,且适用于不同产地的颠茄草,可降低成本,对提升颠茄流浸膏的制备工艺有一定价值。 Objective To comparative study the effectiveness of enzymolysis method and ethanol percolation method used in extracting of effective components in Belladonna herb.Methods Belladonna herb was extracted by cellulase,amylase,pectinase and plant extraction complex enzymes.Compared with ethanol percolation method,the contents of hyoscyamine sulfate and Scopolamine lactone in Belladonna extract were determined by HPLC.Results The complex enzyme was selected as the optimal enzyme.The extraction rate of hyoscyamine sulfate and Scopolamine lactone extracted by the complex enzyme were 95.57%and 94.50%,respectively.The extraction rate of hyoscyamine sulfate and Scopolamine lactone extracted by ethanol percolation method were 85.43%and 85.98%,respectively.The extraction rate of enzymolysis method was better than that of ethanol percolation method.Conclusion The extraction efficiency of belladonna flow extract prepared by enzymatic hydrolysis method is better than that of ethanol percolation method,and it is suitable for belladonna grass from different habitats,which can reduce the cost and is of certain value to improve the preparation process of belladonna flow extract.
