摘要
目的 探讨补肾中药复方对去卵巢骨质疏松模型大鼠皮质骨 (股骨 )生物力学性能的作用及其相关的机制。方法 将 50只 1 0月龄Wistar雌性大鼠随机分为 5组 ,每组 1 0只。正常对照组做假手术 ,其余 4组做卵巢切除术。术后正常饲养 90 d,第 91天开始给药 ,连续用药 90 d,处死 ,测定股骨的骨矿密度、几何尺寸和股骨的生物力学性能。结果 模型大鼠股骨的生物力学性能明显下降 ,骨强度降低 ;股骨干外径变细 ,股骨中段骨皮质面积减少 ,骨矿密度有所下降。补肾中药复方组 (密骨胶囊组和仙灵骨葆组 )大鼠股骨的生物力学性能明显提高 ,骨强度增加 ;股骨中段外径增粗 ,骨皮质面积增加 ,骨矿密度提高。结论 补肾中药复方通过提高去卵巢骨质疏松模型大鼠皮质骨 (股骨 )的宏观结构生物力学应答机能 ,明显改善其生物力学性能。
Objective To probe into the action and active mechanism of 'Traditional Chinese Medicine(TCM) complex prescription of Kidney tonifing' ameliorated biomechanics property of the compact bone in ovariectomized rat. Methods Fifty Wistar female rats aged 10 months were divided randomly into 5 groups: the normal, model, Premarin treated,XianLingGubao treated and MiGuJiaoNang treated, 10 in each group. The normal group was only shamly operated and the other four groups were ovariectomized.The rats after operation were fed normally for ninety days. Ninety one days after operation, the rats were given the medicines for 90 days and then killed. The thighbones were taken out, and the density of bone mineral substance and femoral geometry size and biomechanics capabilities determined.Results Femoral biomechanics property worsened significantly,femoral mechanical strength reduced,femoral diameter diminished,area of the bone cortex decreased and femoral density of bone mineral substance lowered in model group; the biomechanics capabilities of thighbone raised significantly, femoral mechanical strength advanced,femoral diameter augmented,area of the bone cortex aggrandizement and femoral density of bone mineral substance enhanced in 'TCM complex prescription of Kidney tonifing'group (MiGuJiaoNang group and XianLingGuBao group).Conclusions TCM complex prescription of Kidney tonifing could improved the biome chanics property of the os integumentale bone (thighbone) in ovariectomized osteoporosis rat. Its main mechanism of action is that TCM complex prescription of Kidney tonifing could enhance the biomechanics response, accommodate function of macrostructure os integumentale, increase femoral diameter, aggrandize area of the bone cortex and enhance the density of bone mineral substance in ovariectomized rats.
