衢枳壳不同加工饮片对脾虚型大鼠胃肠功能的影响

The effect of Quzhi Fructus Aurantii different processed slices on gastrointestinal function in spleen deficiency rats

目的比较衢枳壳不同加工饮片对脾虚型大鼠胃肠功能的影响。方法SD大鼠随机分为空白组、模型组、阳性对照组(四君子颗粒组),衢枳壳“一体化”饮片低、中、高剂量组,衢枳壳传统饮片低、中、高剂量组,每组各10只(雌雄各半)。脾虚大鼠模型采用苦寒泻下加饥饱失常法复制,连续灌胃给药7 d,给药期间观察大鼠精神状态并记录;末次给药后禁食18 h,用灌胃半固体糊的方法检测各组大鼠的胃排空率以及小肠推进率;用ELISA试剂盒检测大鼠血清中胃泌素(GAS)、肿瘤坏死因子α(TNF-α)、白细胞介素6(IL-6)的含量;血浆中乙酰胆碱(ACh)、血管活性肠肽(VIP)、P物质(SP)、胃动素(MTL)的含量。结果与空白组比较,模型组除SP指标变化无显著性,其余各个指标均具有显著性,其中MTL、GAS、Ach、TNF-α和IL-6含量显著降低(P<0.01),VIP含量显著升高(P<0.01)。各组给药7 d后,与模型组比较,阳性给药组除SP指标无显著性差异,其余指标变化均具有显著性;传统饮片与“一体化”饮片中、高剂量组除SP指标无显著性差异,其余指标变化均具有显著性差异;传统饮片与“一体化”饮片低剂量组,除MTL、GAS、SP无显著性差异,其余指标均具有显著性差异。结论衢枳壳传统饮片与“一体化”饮片对脾虚型大鼠的胃肠功能障碍具有治疗作用,且治疗效果等效。

Objective To compare the effects of different processed pieces of Qu Fructus Aurantii on Gastrointestinal function in Rats with spleen deficiency.Methods 90 SD rats were randomly divided into the blank group,the model group,the positive control group,the low,middle and high dose groups of"integrated"decoction of Qu Fructus Aurantii,and the low,middle and high dose groups of traditional decoction of Qu Fructus Aurantii,with 10 males and females in each group.The rat model of spleen deficiency was established by the method of purgation of bitter cold and abnormal hunger and satiety,and the mental state of rats was observed and recorded during continuous administration for 7 days.After fasting for 18 hours after the last administration,the gastric emptying rate and small intestinal propulsion rate of rats in each group were detected by intragastric administration of semi-solid paste.Enzyme linked immunosorbent assay(ELISA)was used to detect the contents of gastrin(GAS),tumor necrosis factorα(TNF-α)and interleukin 6(IL-6)in serum samples,and acetylcholine(ACh),vasoactive intestinal peptide(VIP),substance P(SP)and motilin(MTL)in plasma samples.Results Compared with the blank group,the contents of MTL,GAS,Ach,TNF-αand IL-6 in the model group decreased significantly,while the content of VIP increased significantly,but the content of SP did not change significantly.After the administration,compared with the model group,there was no significant difference in the SP index in the positive group,there was no significant difference in the SP index in the middle and high dose groups,and there was significant difference in the other indexes except MTL,GAS and SP between the traditional decoction group and the"integrated"decoction group.Conclusion The traditional and"integrated"pieces of Qu Fructus Aurantii have therapeutic effects on gastrointestinal dysfunction in rats with spleen deficiency,and the therapeutic effect is equivalent.
