厚朴麻黄汤抑制哮喘小鼠气道炎症作用机制研究

摘要 目的:探讨厚朴麻黄汤改善哮喘气道炎症的作用机制。方法:将小鼠巨噬细胞随机分成4组(空白对照组、血清对照组、模型组、中药血清组),其中空白对照组不做任何处理;模型组予内毒素处理制备哮喘模型;血清对照组予经生理盐水灌胃小鼠的血清+内毒素处理;中药血清组予经厚朴麻黄汤灌胃小鼠的血清+内毒素处理;采用实时PCR法和Western-blot检测各组COX-2、IL-6及TNF-αmRNA和蛋白表达。结果:与空白组比较,模型组COX-2、IL-6、TNF-a蛋白表达水平明显升高(P﹤0.05);与模型组比较,中药血清组COX-2、IL-6、TNF-a蛋白表达水平明显降低(P﹤0.05),其表达水平受到明显抑制。与模型组比较,中药血清组COX-2、IL-6、TNF-a mRNA相对表达下调,差异有统学意义(P﹤0.05)。结论:内毒素刺激小鼠肺泡巨噬细胞中COX-2、IL-6及TNF-α基因表达,并促进蛋白质表达,厚朴麻黄汤能抑制COX-2、IL-6及TNF-α基因及蛋白质的表达。 Objective:To approach the inhibiting mechanism of Houpu Mahuang decoction on asthma-caused airway inflammation.Methods:Mouse macrophages were randomly divided into four groups(blank control group,serum control group,model group,TCM serum group).Blank control group was not given any treatment.Model group was treated with endotoxin and made into asthma model.Serum control group was treated with serum of mice given normal saline by gavage and endotoxin.TCM serum group was treated with serum of mice given Houpu Mahuang decoction by gavage and endotoxin.The expressions of COX2,IL-6,TNF-α,mRNA and protein were detected in each group by Real-time PCR and Western-blot.Results:Compared with blank control group,the levels of expressions of COX2,IL-6,TNF-αand protein of model group were increased significantly(P<0.05).Compared with model group,the levels of expressions of COX2,IL-6,TNF-a and protein of TCM serum group were decreased significantly(P<0.05),and the expression level was obviously inhibited.Compared with model group,the relative expressions of COX-2,IL-6 and TNF-a mRNA of TCM serum group were down-regulated,and the difference was statistically significant(P<0.05).Conclusion:The endotoxin stimulates the expressions of COX2,IL-6 and TNF-αgene in mouse alveolar macrophages,and promotes the expression of protein.Houpu Mahuang decoction can inhibit the expressions of COX2,IL-6,TNF-αgene and protein.
