水飞蓟宾糖基化衍生物对肝癌细胞凋亡和周期的影响

摘要 目的探讨水飞蓟宾糖基化衍生物对肝癌细胞凋亡和细胞周期的影响。方法将肝癌细胞LIXC-002随机分为空白对照组(Control)、水飞蓟宾糖基化衍生物治疗组(SLB-X2,3、10、30μmol/L)和水飞蓟宾对照组(SLB,30μmol/L),化合物孵育LIXC-002细胞24 h,分别采用流式细胞术和Annexin VFITC/PI法对LIXC-002细胞周期变化及细胞凋亡情况进行检测。结果与SLB组比较,SLB-X2组能够明显提高LIXC-002细胞凋亡率(均P<0.05),而且呈剂量依赖性,在相同浓度下(30μmol/L),SLB-X2的细胞凋亡率是SLB组的2倍。SLB-X2能够将LIXC-002细胞阻滞在G2/M期,且作用强于SLB。结论SLB-X2能够通过促进肝癌细胞凋亡,使肝癌细胞周期阻滞在G2/M期,发挥抗肝癌作用。 Objective To investigate the effects of silymarin glycosylated derivatives on apoptosis and cell cycle of hepatocellular carcinoma cells LIXC-002.Methods The cells were randomly divided into blank control group(Control),silymarin glycosylated derivative treatment group(SLB-X2,3μmol/L,10μmol/L,30μmol/L)and silyma-rin control group(SLB,30μmol/L),and the compounds were incubated in LIXC-002 cells for 24 hours,and then LIXC-002 cell cycle changes and apoptosis were detected by flow cytometry and Annexin VFITC/PI method,respec-tively.Results Compared with SLB group,SLB-X2 group could significantly increase the apoptosis rate of LIXC-002 cells(P<0.05),and the apoptosis rate of SLB-X2 was twice as high as that of SLB group at the same concentration(30μmol/L);in addition,SLB-X2 could block LIXC-002 cells in G2/M phase,and its effect was stron-ger than that of SLB.Conclusions SLB-X2 can exert anti-hepatocellular carcinoma effects by promoting apoptosis of hepatocellular carcinoma cells and blocking the hepatocellular carcinoma cell cycle in the G2/M phase.
