益脑通络方对急性血瘀合并MCAO/R大鼠血液流变学及凝血功能的影响

Effects of Yinao Tongluo Formula on hemorheology and coagulation function in rats with acute blood stasis combined with MCAO/R model

摘要 目的:探讨益脑通络方对急性血瘀合并大脑中动脉闭塞/再灌注(MCAO/R)模型大鼠血液流变学及凝血功能的影响。方法:选取SPF级雄性SD大鼠140只,随机分为假手术组、血瘀组、血瘀缺血组、阳性药物组及益脑通络方低、中、高剂量组,每组20只。连续灌胃7 d后,除假手术组外各组均制备急性血瘀模型;末次给药30 min后,除血瘀组及假手术组外各组合并制备MCAO/R模型模型,假手术和血瘀组仅分离颈总动脉不插入线栓阻断血流。再灌注24 h后,各组随机选取8只大鼠取脑,剩余大鼠腹主动脉取血,对脑含水量、脑梗死体积百分比、全血黏度、血浆黏度、凝血四项、红细胞压积及红细胞相关参数进行检测。结果:与假手术组比较,血瘀缺血组大鼠脑含水量显著增加(P<0.05),脑梗死体积百分比显著增加(P<0.05),血液在低、中、高切变率下全血黏度及血浆黏度显著上升(P<0.05),红细胞压积、聚集指数、刚性指数显著升高(P<0.01,P<0.05),红细胞变形指数、电泳指数显著降低(P<0.01,P<0.05),APTT、TT、PT时间显著缩短(P<0.05),FIB含量显著增加(P<0.01);与血瘀缺血组比较,益脑通络方中、高剂量组脑含水量显著降低(P<0.01),脑梗死体积百分比显著降低(P<0.05,P<0.01),血液在高、中、低切变率下的全血黏度及血浆黏度均显著下降(P<0.01,P<0.05),红细胞压积、聚集指数、刚性指数显著降低(P<0.01,P<0.05),红细胞变形指数、电泳指数显著升高(P<0.01,P<0.05),APTT、TT、PT时间显著延长(P<0.01,P<0.05),同时FIB含量显著降低(P<0.05)。结论:益脑通络方可以显著改善脑水肿,缩小脑梗死面积,降低血液黏稠度,改善血液黏滞状态。 Objective:To investigate the effects of Yinao Tongluo Formula on hemorheology and coagulation function in rats with acute blood stasis combined with MCAO/R model.Methods:A total of 140 SPF male SD rats were randomly divided into sham operation group,blood stasis group,blood stasis ischemia group,positive drug group and low,medium and high doses Yinao Tongluo Formula groups.After continuous gavage for 7 days,acute blood stasis models were prepared in all groups except sham operation group.Thirty minutes after the last administration,MCAO/R models were prepared in all groups except blood stasis group and sham operation group.In sham operation and blood stasis group,only common carotid artery was separated without inserting thread plug to block blood flow.24 hours after reperfusion,the brains of 8 rats in each group were randomly selected,and the blood was taken from abdominal aorta of the remaining rats,and the indexes of brain tissue water content,cerebral infarction volume,whole blood viscosity,plasma viscosity,coagulation,hematocrit and erythrocyte factors were detected.Results:Compared with the sham operation group,the amount of cerebral water content in the blood stasis ischemia group were increased significantly(P<0.01),the percentage of cerebral infarction were increased significantly(P<0.05),blood in low,medium,high-cut rate,whole blood viscosity and plasma viscosity was significantly increased(P<0.05),red blood cell compaction,red blood cell aggregation index,rigid index were significantly increased(P<0.01,P<0.05),red blood cell deformation index,electrophoresis index were significantly decreased(P<0.01,P<0.05),APTT,TT,PT time were significantly shorten(P<0.05),and the FIB content were increased significantly(P<0.01).Compared with the blood stasis ischemia group,the brain water content ratio in the medium and high doses Yinao Tongluo Formula were decreased significantly(P<0.01),the volume percentage of cerebral infarction were decreased significantly(P<0.05,P<0.01),the whole blood viscosity and plasma viscosity of blood at high,medium and low shear rates were decreased significantly(P<0.01,P<0.05),the red blood cell compaction,the red blood cell aggregation index,the rigid index were significantly decreased(P<0.01,P<0.05),the red blood cell deformation index,the electrophoresis index was significantly increased(P<0.01,P<0.05),APTT,TT,PT time were significantly prolonged(P<0.01,P<0.05),and the FIB content was significantly reduced(P<0.05).Conclusion:Yinao Tongluo Formula can significantly improve cerebral edema,shrink the cerebral infarction area,reduce blood viscosity,improve blood viscous state.
