基于转录组学探讨四君子汤增强顺铂抗胃癌作用的机制 被引量:1

Mechanism of Sijunzi Decoction on enhancing the anti-gastric cancer effect of cisplatin based on transcriptomics

摘要 目的:采用转录组学方法探讨四君子汤增强顺铂抗胃癌的作用及其机制。方法:建立人胃癌裸鼠移植瘤模型,随机分为模型组、顺铂组、四君子汤联合顺铂组,测量各组裸鼠体质量、肿瘤体积和瘤重。通过转录组测序分析筛选差异基因并对差异基因进行功能注释与富集分析。结果:与顺铂组比较,四君子汤联合顺铂组肿瘤体积、瘤重、瘤重指数均显著降低(P<0.01,P<0.05),顺铂组的抑瘤率为25.99%,四君子汤联合顺铂组的抑瘤率为79.14%。四君子汤联合顺铂组与顺铂组共有31个差异表达基因,涉及PI3K/Akt、ErbB等信号通路。结论:四君子汤可以增强顺铂抗胃癌作用,其作用机制可能与PI3K/Akt、ErbB等信号通路有关。 Objective:To investigate the mechanism of Sijunzi Decoction on enhancing anti-gastric cancer effect of cisplatin by transcriptomics technique.Methods:Nude mice were subcutaneously transplanted with SGC-7901 cells,then the mice were randomly divided into three groups:model group,cisplatin group,Sijunzi Decoction-cisplatin combined group.The body weight,tumor volume and tumor weight of nude mice were measured.RNA-Seq was performed to analyze the mRNA expression profiles of tumor tissues.In addition,functional annotation and enrichment analysis of differential genes were performed.Results:Compared with the cisplatin group,the tumor volume,tumor weight and tumor weight index of Sijunzi Decoction-cisplatin combined group were significantly decreased(P<0.01,P<0.05),tumor inhibition rate of cisplatin group and Sijunzi Decoction-cisplatin combined group were 25.99%and 79.14%.Transcriptomic sequencing data showed that 31 differently expressed genes in Sijunzi Decoction-cisplatin combined group compared with cisplatin group were mainly enriched in the PI3K/Akt and ErbB signaling pathways.Conclusion:Sijunzi Decoction enhances anti-gastric cancer effect of cisplatin and the mechanisms of action of Sijunzi Decoction may be related to the regulation of PI3K/Akt and ErbB signaling pathways.
