摘要
试验旨在采用超声波提取全缘金粟兰总黄酮,综合单因素试验和正交试验结果优选其最佳提取条件,研究全缘金粟兰地上与地下总黄酮对1,1-二苯基-2-三硝基苯肼(DPPH)自由基及羟自由基的清除率。结果表明,全缘金粟兰总黄酮的最佳提取工艺条件为乙醇体积分数45%、超声时间15 min、液料比45 mL/g,在此条件下全缘金粟兰总黄酮提取率为7.68%。全缘金粟兰地上部位与地下部位对DPPH自由基及羟自由基的清除能力呈一定的量效关系,在0.425、0.460 g/mL时对DPPH自由基的清除率高达98.3%和98.2%,二者对于羟自由基的清除率在质量浓度为0.085~0.450 g/mL的范围内呈现一定的量效关系,但是同等浓度下二者对DPPH自由基及羟自由基的清除能力相反。研究表明,试验结果可为全缘金粟兰物质成分基础研究以及药材使用部位的选择提供研究方向和依据。
The experiment aims to use ultrasound to extract total flavonoids from Cymbidium holostegius.Based on the results of single factor experiments and orthogonal experiments,the optimal extraction conditions are optimized.The antioxidant activity of total flavonoids from Cymbidium holostegius is studied by clearance rates of DPPH free radical and OH free radical by both aboveground and underground flavonoids.The experimental results showed that the optimal extraction process conditions for total flavonoids from Cymbidium holostegius were ethanol volume fraction of 45%,ultrasound time of 15 min,and liquid-solid ratio of 45 mL/g.Under these conditions,the extraction rate of total flavonoids from Cymbidium holostegius was 7.68%.There was a certain dose-effect relationship between the above ground and underground parts of Cymbidium holostegius,and the clearance rates of DPPH free radical and OH free radical are as high as 98.3%and 98.2%at 0.425,0.460 g/mL,respectively.The clearance rates of 0H free radical between the two parts show a certain dose-effect relationship in the mass concentration range of 0.085~0.450 g/mL.However,at the same concentration,the two have opposite scavenging abilities for DPPH free radical and OH free radical.The study indicates that the experimental results could provide the research direction and basis for the basic research of the material composition and the selection of the medicinal parts of Cymbidium holostegius.
