Mito-Tempo修复过氧化氢诱导的猪卵母细胞线粒体损伤机制研究 被引量:1

摘要 本研究探讨线粒体靶向抗氧化剂Mito-Tempo对过氧化氢诱导猪卵母细胞线粒体损伤修复的影响。在猪卵母细胞体外成熟过程中添加10 mmol·L^(-1)过氧化氢或过氧化氢+0.1μmol·L^(-1)Mito-Tempo,检测卵丘细胞扩散、卵母细胞成熟及线粒体功能相关因子,观察孤雌胚胎发育情况。结果表明,与过氧化氢组相比,添加Mito-Tempo可显著提高卵丘细胞扩散指数,显著降低Mito SOX水平,提高膜电位及线粒体功能基因表达水平,显著改善猪卵母细胞体外成熟率及孤雌胚胎发育能力(P<0.05)。证明Mito-Tempo对过氧化氢诱导的猪卵母细胞线粒体损伤具有修复功能。 To investigate the effect of Mito-Tempo on H2O2-induced mitochondrial dysfunction in porcine oocytes,supplemented 10 mmol·L^(-1) H_(2)O_(2) and/or 0.1μmol·L^(-1) Mito-Tempo to oocyte maturation medium,evaluated the cumulus cell expansion,oocyte maturation and mitochondria-relative factors,and obsened subsequent embryo development competence after parthenogenetic activation.The results showed that Mito-Tempo significantly improved cumulus expansion index compared to the H_(2)O_(2) group(P<0.05),Mito-Tempo addition significantly reduced the level of MitoSOX,and increased the mitochondrial membrane potential and mitochondria-relative gene expression(P<0.05),additionally,Mito-Tempo significantly improved porcine oocyte maturation rate in vitro and embryo developmental competence(P<0.05).In conclusion,Mito-Tempo could rescue the mitochondrial dysfunction from H_(2)O_(2)-induced damage during porcine in vitro oocyte maturation.
