摘要
目的探讨细胞分裂周期相关蛋白3(CDCA3)在上皮性卵巢癌中的表达及其对癌细胞增殖、侵袭和迁移的影响。方法收集于本院进行手术切除的50例患者的卵巢癌组织和30例患者的正常卵巢组织,选取A2780、Hey、CAOV3、SKOV3细胞进行研究。RT-PCR检测组织和细胞中CDCA3 mRNA表达水平,Western blot检测组织和细胞中CDCA3蛋白表达水平。将CAOV3细胞分为沉默组和Control组,沉默组转染CDCA3 siRNA,Control组转染阴性对照序列;将Hey细胞分为过表达组和对照组,过表达组用CAOV3慢病毒感染,对照组用空载体慢病毒感染。CCK-8检测细胞活性;Transwell实验检测细胞侵袭和迁移。结果与正常卵巢组织比较,上皮性卵巢癌组织中CDCA3 mRNA和蛋白表达水平较高(P<0.05);在A2780、Hey、CAOV3、SKOV3上皮性卵巢癌细胞系中,CAOV3 mRNA和蛋白表达水平最高,而Hey最低。沉默CDCA3后CAOV3细胞CDCA3蛋白表达水平、细胞增殖活力、侵袭和迁移能力降低(P<0.05),Cyclin D1、MMP-9蛋白表达水平降低(P<0.05),而E-cadherin蛋白表达水平升高(P<0.05)。过表达CDCA3后Hey细胞CDCA3蛋白表达水平、细胞增殖活力、侵袭和迁移能力升高(P<0.05),Cyclin D1、MMP-9蛋白表达水平升高(P<0.05),而E-cadherin蛋白表达水平降低(P<0.05)。结论CDCA3在上皮性卵巢癌中高表达,能够促进上皮性卵巢癌细胞的增殖、侵袭和迁移。
Objective To investigate the expression of cell division cycle associated protein 3(CDCA3)in epithelial ovarian cancer and its effect on the proliferation,invasion and migration of cancer cells.Methods The ovarian cancer tissue of 50 patients and the normal ovarian tissue of 30 patients who underwent surgical resection in our hospital were collected,and A2780,Hey,CAOV3,SKOV3 cells were selected for research.RT-PCR was used to detect the expression level of CDCA3 mRNA in tissues and cells,and Western blot was used to detect the expression level of CDCA3 protein in tissues and cells.CAOV3 cells were divided into the silence group and the Control group,the silence group was transfected with CDCA3 siRNA,while the Control group was transfected with negative control sequence.Hey cells were divided into the overexpression group and the control group,the overexpression group was infected with CAOV3 lentivirus,and the con-trol group was infected with empty vector lentivirus.Cell viability was detected by CCK-8;and the cell invasion and migration were detected by Transwell assay.Results Compared with normal ovarian tissue,the expression levels of CDCA3 mRNA and CDCA3 protein in epithelial ovarian cancer tissue were higher(P<0.05).Among the epithelial ovarian cancer cell lines A2780,Hey,CAOV3,and SKOV3,the expres-sion levels of CAOV3 mRNA and CDCA3 protein were the highest,while the expression levels of Hey were the lowest.After CDCA3 was silenced,the expression level of CDCA3 protein,cell proliferation activity,invasive and migration ability of CAOV3 cells decreased(P<0.05),the expression levels of Cyclin D1 and MMP-9 protein decreased(P<0.05),while the expression level of E-cadherin protein increased(P<0.05).After CDCA3 was overexpressed,the expression level of CDCA3 protein,cell proliferation activity,invasive and migration ability of Hey cells increased(P<0.05),the expression levels of Cyclin D1 and MMP-9 protein increased(P<0.05),while the expression level of E-cadherin protein decreased(P<0.05).Conclusion CDCA3 is highly expressed in epithelial ovarian cancer and can promote the proliferation,invasion and migration of epithelial ovarian cancer cells.
