鸡增殖型与肥大型软骨细胞差异表达基因筛选

Screening of Differentially Expression Genes in Chicken Proliferative and Hypertrophic Chondrocytes

摘要 为研究家禽软骨细胞分化过程中的差异基因,寻找家禽胫骨软骨发育不良发生的相关基因和通路,研究基于GEO数据库下载家禽软骨细胞分化的基因表达谱芯片数据进行生物信息学分析。结果显示:共筛选出292个差异基因,其中40个上调,252个下调;差异基因的GO富集主要在细胞成分活动的调节、趋化性、细胞间黏附和细胞黏附分子结合等过程,KEGG信号通路分析主要在ECM受体相互作用、黏着斑等通路富集;PPI网络分析筛选出APP、SPP1、TF和LAMB14个核心基因,并结合RT-PCR技术对核心基因进行表达谱验证,与预测的结果一致,均表现在增殖型软骨细胞中表达下调。研究表明,从GEO18568数据中分析筛选出的4个核心基因,在增殖型软骨细胞分化为肥大型软骨细胞过程中均发挥正向调控作用。 This work was to investigate the differential genes in the differentiation process of poultry chondrocytes,and to find related genes and pathways for the occurrence of tibial chondrodysplasia.The gene expression profile chip of poultry chondrocyte differentiation based on GEO database is used for bioinformatics analysis.The results showed that it was a total of 292 differential genes were screened,of which 40 were up-regulated and 252 were down-regulated through bioinformatics analysis.The GO enrichment of differential genes was mainly in regulation of cellular component movement,chemotaxis,cell-cell adherens junction and cell adhesion molecule binding.The analysis of KEGG signaling pathway was mainly enriched in ECM receptor interaction,focal adhesion.PPI network analysis screened out 4 core genes:APP,SPP1,TF and LAMB1.It was used by RT-PCR technology to verify the expression profile of the core genes,which was consistent with the predicted results,and the expression was down-regulated in the proliferative chondrocytes.The results of this study indicated that APP,SPP1,TF and LAMB1 played an important role in the process of cartilage differentiation.The results indicated that 4 core genes which were analyzed and screened from GEO18568,all played a positive regulatory role in the differentiation of proliferative chondrocytes into hypertrophic chondrocytes.
