一株基因Ⅴ型鸡呼肠孤病毒的分离与鉴定

摘要 为确定山东省某商品肉鸡场发病病原,掌握其遗传进化特征,研究通过采集发病鸡场患鸡肌腱进行病原分离,经RT-PCR、序列分析等方法进行鉴定。结果显示:从发病鸡群中分离到一株呼肠孤病毒,该分离株(YT株)与ARV基因V型参考株K100557处同一进化分支,核苷酸和氨基酸相似性达96.0%,氨基酸序列仅存在13个突变位点;而与疫苗株S1133的核苷酸和氨基酸相似性仅为54.4%和49.2%,氨基酸序列存在167个突变位点。研究表明该鸡场发病病原为基因V型鸡呼肠孤病毒,为制定综合防控措施提供参考。 To determine the pathogenic agent of a commercial broiler farm in Shandong Province and to grasp its genetic evolutionary characteristics,the study was conducted by collecting tendons of affected chickens from the affected farm for pathogen isolation and identified by RT-PCR and sequence analysis.The results showed that a strain of eutherian virus was isolated from the affected chickens,and the isolate(YT strain)was in the same evolutionary branch as the ARV gene type V reference strain K100557,with the highest nucleotide and amino acid homology of 96.0%,and only 13 mutation sites in amino acid sequence;While the nucleotide and amino acid homology with the vaccine strain S1133 were only 54.4% and 49.2%,and the amino acid sequence had 167 amino acid mutations.The study showed that the pathogen of this chicken farm was genotype V Avian Orthoreovirus,which provided a reference for the development of integrated prevention and control measures.
