摘要
为了寻找有效抗冠状病毒感染的治疗靶点,用CCK-8法和结晶紫法检测VPS34蛋白抑制剂PIKⅢ对细胞的敏感性,通过CCK-8法检测PIKⅢ抗病毒活性,通过病毒滴度及RT-PCR检测PIKⅢ对冠状病毒复制的影响。结果显示,PIKⅢ可以抑制多种冠状病毒对细胞的杀伤作用,提高细胞活性并且减少冠状病毒复制。VPS34蛋白抑制剂PIKⅢ对SARS-CoV-2、PEDV和TGEV 3种冠状病毒感染具有抑制作用,可以减少病毒复制,PIKⅢ可能是一种有效的抗冠状病毒药物。
In order to find effective therapeutic targets against coronavirus infection,in this study,CCK-8 and crystal violet methods were used to detect the sensitivity of VPS34 protein inhibitor PIKⅢto cells,CCK-8 method was used to detect the antiviral activity of PIKⅢ,RT-PCR and virus titer were used to detect the effect of PIKⅢon the replication of coronavirus.The results showed that PIKⅢcould inhibit the killing effect of various coronaviruses in cells,improve cell activity and reduce the replication of coronaviruses.The results showed that VPS34 protein inhibitor PIKⅢhas inhibitory effect on SARS CoV-2,PEDV and TGEV.PIKⅢcan reduce virus replication.In conclusion,PIKⅢmaybe an effective anti-coronavirus drug.
