云南部分地区家畜寄生蜱携带病原情况调查

Investigation on Pathogens Carried by Ticks on Livestock in Partial Areas of Yunnan

摘要 为了解云南省部分地区家畜寄生蜱的携带病原(贝氏柯克斯体、伯氏疏螺旋体、回归热螺旋体)情况,从云南玉溪市、昆明市、楚雄市、普洱市的牛、羊体表收集455只蜱,形态学及分子生物学鉴定为微小扇头蜱,通过贝氏柯克斯体ComⅠ基因、伯氏疏螺旋体5S-23S rRNA基因、回归热螺旋体16S rRNA基因对这些蜱传病原进行检测,并对阳性扩增产物进行测序、序列比对,并构建进化树。结果发现检测到47份蜱虫样品呈贝氏柯克斯体阳性,2份蜱虫样品呈回归热螺旋体阳性,序列比对发现检测到的回归热螺旋体与色勒疏螺旋体序列高度一致,但未检测到伯氏疏螺旋体。发现云南省贝氏柯克斯体呈散发流行状态,首次在云南发现并报道了色勒疏螺旋体的存在和传播。 In order to reveal the prevalence of pathogens carried by ticks on livestock(Coxsiella burnetii,Borrelia burgdorferi,and Spirochaeta recurrentis)in partial areas of Yunnan province,in this study,455 ticks were collected from cattle and sheep in Yuxi city,Kunming city,Chuxiong city and Pu'er city of Yunnan province.The ticks were identified as Rhipicephalus microplus by using morphology and molecular biology methods.In this study,ComⅠgene of Coxsiella burnetii,5S-23S rRNA of Borrelia burgdorferi,and 16S rRNA of Spirochaeta recurrentis were used for detection of these pathogens.DNA sequencing and alignment were performed,and the evolutionary tree was constructed.The results showed that 47 tick samples were positive for Coxsiella burnetii,and 2 tick samples were positive for Spirochaeta recurrentis.Sequence alignment showed that the Spirochaeta recurrentis detected in this study showed highly consistent with Borrelia theileri,but Borrelia burgdorferi was not detected in this study.This study found that Coxsiella burnetii was in sporadic epidemic state in Yunnan province,and reported the existence and transmission of Borrelia theileri in Yunnan province for the first time.
