CCL28在子宫内膜异位症中的作用机制研究

摘要 目的探讨C-C基序趋化因子配体28(CCL28)在子宫内膜异位症(EM)中的表达及可能机制。方法比较15例卵巢子宫内膜异位组织及15例在位内膜组织中CCL28mRNA及蛋白表达水平。将携带CCL28全基因的慢病毒(CCL28组)及空载体病毒(Emptyvector组)、CCL28-siRNA(CCL28-siRNA组)及其对照基因(CON组)分别转入人子宫内膜基质细胞HESs。检测各组CCL28 mRNA及蛋白表达水平、上皮间质转化(EMT)相关指标(E-cadherin、N-cadherin、Snail-1、Vimentin)蛋白表达水平、细胞活性、细胞侵袭和迁移。结果卵巢子宫内膜异位组织CCL28 mRNA和蛋白表达水平均高于在位内膜组织(P<0.05)。与Emptyvector组比较,CCL28组CCL28mRNA及蛋白表达水平、细胞增殖力、侵袭力、迁移力增高(P<0.05)。与Emptyvector组比较,CCL28组E-cadherin蛋白表达水平较低,而N-cadherin、Snail-1和Vimentin蛋白表达水平较高(P<0.05)。与CON组比较,CCL28-siRNA组CCL28 mRNA及蛋白表达水平、细胞增殖力、侵袭力、迁移力降低(P<0.05)。与CON组比较,CCL28-siRNA组E-cadherin蛋白表达水平较高,而N-cadherin、Snail-1和Vimentin蛋白表达水平较低(P<0.05)。结论CCL28可能通过影响细胞增殖、侵袭、迁移和EMT参与EM发生发展。 Objective To investigate the expression of C-C motif chemokine ligand 28(CCL28)in endometriosis(EM)and its possible mechanism.Methods The expression levels of CCL28 mRNA and protein in 15 cases of ovarian endometriosis and 15 cases of eutopic endometrial tissue were compared.The lentivirus(CCL28 group),empty vector virus(Empty vector group),CCL28-siRNA(CCL28-siRNA group)and its control gene(CON group)carrying the full CCL28 gene were transferred into human endometrial stromal cell HESs,respectively.CCL28 mRNA and protein expression level,expression level of epithelial mesenchymal transformation(EMT)related indicators(E-cadherin,N-cadherin,Snail-1,Vimentin)protein,cell activity,cell invasion and migration were detected in each group.Results The expression levels of CCL28 mRNA and protein in ovarian endometriosis tissue were higher than those in eutopic endometrial tissue(P<0.05).Compared with the Empty vector group,the CCL28 mRNA and protein expression levels,cell proliferation,invasion,and migration of the CCL28 group were increased(P<0.05).Compared with the Empty vector group,the E-cadherin protein levels was lower in the CCL28 group,while N-cadherin,Snail-1 and Vimentin protein levels were higher in the CCL28 group(P<0.05).Compared with the CON group,the CCL28-siRNA group had lower CCL28 mRNA and protein expression levels,cell proliferation,invasion,and migration(P<0.05).Compared with CON group,E-cadherin protein expression level was higher in CCL28-siRNA group,while N-cadherin,Snail-1 and Vimentin protein expression levels were lower(P<0.05).Conclusion CCL28 may be involved in the development of EM by affecting cell proliferation,invasion,migration and EMT.
