生存相关DLEU1与HPV感染阳性宫颈癌关系研究

Survival related⁃DLEU1 is associated with HPV infection status and serves as a biomarker in HPV-infected cervical cancer

目的 探讨淋巴细胞白血病中lncRNA缺失型1(DLEU1)是否与人乳头瘤病毒(HPV)感染相关以及DLEU1在HPV阳性(HPV+)宫颈癌(CC)患者中的临床价值。方法 选择2016年1月~2017年12月接受治疗的CC患者128例作为宫颈癌组,同期行子宫切除术的子宫肌瘤患者99例作为对照组,通过受试者工作特征分析评估DLEU1从对照中筛选CC患者的能力以及区分不同HPV感染状态个体的能力,利用Kaplan-Meier生存分析和Cox回归分析评估DLEU1与CC患者生存预后的关系。结果 DLEU1表达在CC组织和细胞系中显著上调,特别是在HPV+的组织和细胞系中。DLEU1在区分CC患者以及区分HPV+和HPV阴性(HPV-)CC患者方面具有较高的诊断价值,并对HPV+对照具有一定的筛选能力。结论 DLEU1与CC患者HPV感染相关,与总CC患者和HPV+CC患者的生存预后相关,并可独立预测CC患者的预后,DLEU1可作为HPV+CC患者的诊断和预后生物标志物。

Objective To investigate whether lncRNA deleted in lymphocytic leukemia 1(DLEU1)was associated with HPV in⁃fection,and explore the clinical significance of DLEU1 in HPV-infected cervical cancer(CC)patients.Methods From January 2016 to De⁃cember 2017,128 patients with CC treated in Weifang People's Hospital were selected as the cervical cancer group,and 99 patients with hys⁃teromyoma who underwent hysterectomy during the same period were selected as the control group.The ability of DLEU1 to screen CC patients from controls and to differentiate individuals with different HPV infection status was evaluated by the receiver operating characteristic analysis.The association of DLEU1 with the survival prognosis of CC patients was assessed by Kaplan⁃Meier survival analysis.Results DLEU1 expres⁃sion was significantly upregulated in CC tissues and cell lines,especially increased in HPV-positive cases.Additionally,DLEU1 had a high diagnostic value in distinguishing CC patients,differentiating between HPV-positive and HPV-negative CC patients,and had a certain ability to screen HPV-positive controls.DLEU1 was correlated with HPV infection in CC patients.Moreover,the association of DLEU1 with survival prognosis was found in total CC patients and HPV-positive CC patients.Most of the molecules interacting with DLEU1 were microRNAs.Con⁃clusion Abnormal DLEU1 expression is associated with HPV infection,and may function as a diagnostic and prognostic biomarker for HPV-positive CC patients.
