基于ISSR与SCoT分子标记的赤苍藤种质遗传多样性分析及DNA指纹图谱构建 被引量:2

Genetic Diversity Analysis and DNA Fingerprinting of E.scandens Germplasm Based on ISSR and SCoT Molecular Markers

摘要 为了选育赤苍藤优良品种,本研究利用ISSR与SCoT分子标记技术对24份赤苍藤种质资源进行遗传多样性分析。结果表明,11条ISSR引物共扩增出91个条带,其中多态性带条73个,平均每条引物扩增多态性条带6.6个;10条SCoT引物共扩增出102个条带,其中多态性条带86个,平均每条引物扩增多态性条带8.6个。基于ISSR与SCoT标记的遗传相似系数的范围分别是0.5604~0.8571和0.5294~0.8824,说明赤苍藤种内具有较为丰富的遗传变异,其遗传多样性的表现与地理位置的相关性不显著。ISSR与SCoT标记分别在遗传相似系数0.72和0.69处可将24份赤苍藤种质聚为3类,两种分子标记的聚类结果基本相同。此外,在此研究基础上通过2条ISSR引物组合(UBC824,UBC827)和2条SCoT引物组合(SCoT-2,SCoT-27)分别构建了赤苍藤种质的DNA指纹图谱。本研究为赤苍藤种质资源的保护与利用、品种的鉴定与分类、选育与改良提供了理论依据。 In order to breed superior variety of Erythropalum scandens Blume,ISSR and SCoT markers were used to analyze the genetic diversity of 24 germplasms in this study.The results showed that 91 bands were amplified by 11 ISSR primers,including 73 polymorphic bands,with an average of 6.6 polymorphic bands.102 bands were amplified by 10 SCoT primers,including 86 polymorphic bands,with an average of 8.6 polymorphic bands.The genetic similarity coefficients ranged from 0.5604 to 0.8571 and 0.5294 to 0.8824 based on ISSR and SCoT markers,indicating that there were abundant genetic variations within the species,the correlation between genetic diversity and geographical location was not significant.24 germplasms could be divided into 3 groups at the genetic similarity coefficients of 0.72 and 0.69 base on ISSR and SCoT markers.The clustering results of the two markers are roughly the same.In addition,two ISSR primers(UBC824,UBC827)and two SCoT primers(SCoT-2,SCoT-27)were used to construct the DNA fingerprints of Erythropalum scandens Blume.The study result provides a theoretical basis for the protection and utilization of germplasm resources,identification and classification of varieties,and breeding of Erythropalum scandens Blume.
