HPLC法测定盐酸催化甘蔗渣水解产物中的糖组分

摘要 建立高效液相色谱法测定甘蔗渣盐酸催化水解液中的糖组分。采用Aminex HPX-87P色谱柱(300mm×7.8 mm,粒径9μm,交联率8%),以超纯水为流动相,流速为0.60 mL/min,柱温75℃,进样量10μL。以峰面积定量,结果表明:D-葡萄糖、D-木糖、D-半乳糖、L-阿拉伯糖分别在0.5000~5.000 mg/mL(R=0.9996),2.000~20.00 mg/mL (R=0.9999),0.0260~0.2600 mg/mL (R=0.9991),0.2000~2.000 mg/mL(R=0.9996)范围内有良好的线性关系,平均回收率(n=9)分别为99.05%、99.53%,98.50%、98.63%,重复性实验中,4种糖含量的RSD(n=5)分别为0.13%、0.05%、0.86%、0.46%。本试验方法效率高、重现性及回收率好,可有效应用于甘蔗渣盐酸水解液糖组分的检测。 HPLC was established to determine the sugar components in the acid hydrochloride catalyzed hydrolysate of bagasse.Methods Aminex HPX-87P column(300 mm×7.8 mm,particle size 9μm,8%)was adopted,ultrapure water was used as mobile phase at a flow rate of 0.60 mL/min,the column temperature was set at 80℃,and the injection volume was 10μL.By the peak area,the results showed that the D-Glucose,D-Xylose,D-Galactose,and L-Arabinose had good linearity in the range of 0.2000~2.000 mg/mL(R=0.9996),2.000~20.00 mg/mL(R=0.9999),0.0260~0.2600 mg/mL(R=0.9991),0.2000~2.000 mg/mL(R=0.9996),respectively;The average recovery rate(n=9)were 99.05%,99.53%,98.50%,98.63%,respectively.In the repeatability experiments,the RSD(n=5)for the four sugar contents were 0.13%,0.05%,0.86%,and 0.46%,respectively.The method had high efficiency,good repeatability and good recovery rate,and can be effectively used for sugarcane bagasse hydrochloric acid hydrolysate detection.
