管花肉苁蓉饮片中主要苯乙醇苷近红外快检技术的建立及应用 被引量:3

Establishment and application of near-infrared rapid detection technology for main phenylethanoid glycosides in Cistanche tubolosa decoction pieces

摘要 利用近红外快检技术,建立了快速测定管花肉苁蓉饮片中4种主要苯乙醇苷含量的方法。以60批管花肉苁蓉饮片为研究对象,高效液相色谱法(HPLC)测定管花肉苁蓉饮片中4种主要苯乙醇苷(松果菊苷、管花苷A、毛蕊花糖苷、异毛蕊花糖苷)的含量作为参考值,采用近红外光谱法(NIRS)结合偏最小二乘法(PLS)建立管花肉苁蓉饮片中4种苯乙醇苷含量的定量分析模型。结果表明:该定量分析模型中4种主要苯乙醇苷的最佳光谱预处理方法分别为二阶导数法(2nd Der)+多元散射校正法(MSC)+SG平滑法(SG Smth), 2nd Der+SG Smth, 2nd Der+MSC+SG Smth, 2nd Der+MSC+SG Smth;校正决定系数(Rc2)分别为0.9631, 0.9965, 0.9839, 0.9467;预测决定系数(Rp2)分别为0.8472, 0.8589, 0.8613, 0.8109;校正均方根偏差(RMSEC)分别为0.3236, 0.0058, 0.0581, 0.0237;预测均方根偏差(RMSEP)分别为0.6656,0.0383, 0.1748, 0.0457。采用NIRS法测定管花肉苁蓉饮片中4种主要苯乙醇苷含量与HPLC测定结果无显著差异。 A rapid method for the determination of four main phenylethanoid glycosides in Cistanche tubolosa decoction pieces was established based on near-infrared rapid detection technology.Taking 60 batches of Cistanche tubolosa decoction pieces as the samples,the contents of four major phenylethanoid glycosides(echinosides,tubuloside A,acteoside and isoacteoside)in Cistanche tubolosa decoction pieces determined by high performance liquid chromatography(HPLC)were taken as reference values.The near-infrared spectroscopy(NIRS)combined with partial least squares(PLS)method was used to establish a quantitative analysis model for predicting the contents of four main phenylethanoid glycosides in Cistanche tubolosa decoction pieces.The results suggested that the optimal spectral pretreatment methods for echinosides,tubuloside A,acteoside and isoacteoside were second derivative(2nd Der)+multiplicative scatter correction(MSC)+savitzky-golay smoothing(SG Smth),2nd Der+SG Smth,2nd Der+MSC+SG Smth and 2nd Der+MSC+SG Smth in this quantitative analysis model.The correlation coefficients of set(R_c~2)and predictation set(R_p~2)were 0.9631,0.9965,0.9839 and 0.9467,and 0.8472,0.8589,0.8613 and 0.8109,respectively.The root mean square errors of calibration(RMSEC)and predication(RMSEP)were 0.3236,0.0058,0.0581 and 0.0237,and 0.6656,0.0383,0.1748 and 0.0457,respectively.The contents of four main phenylethanoid glycosides in Cistanche tubulosa decoction pieces determined by the established NIRS method were not significantly different from those determined by HPLC.
