Simultaneous determination of ten nucleosides in Wuling Capsules/Wuling fungus powder by UPLC-MS/MS

摘要 目的建立UPLC-MS/MS法同时测定乌灵胶囊/乌灵菌粉中腺苷、胞苷、鸟苷、肌苷、尿苷、脱氧鸟苷、胞嘧啶核苷酸、尿嘧啶核苷酸、鸟嘌呤核苷酸、腺嘌呤核苷酸的含量。方法该药物水提液的分析采用Thermo Hypercard色谱柱(2.1 mm×100 mm,5μm);流动相水(含2 mmol/L乙酸铵、0.06%乙二胺)-乙腈(含2 mmol/L乙酸铵、0.06%乙二胺),梯度洗脱;体积流量0.6 mL/min;柱温40℃;电喷雾离子源;负离子扫描;多反应监测模式。结果10种成分在各自范围内线性关系良好(R^(2)>0.9900),平均加样回收率85.52%~104.21%,RSD 0.79%~4.48%。结论该方法简便可行,重复性好,可用于乌灵胶囊/乌灵菌粉的质量控制。 AIM To establish a UPLC-MS/MS method for the simultaneous content determination of adenosine,cytidine,guanosine,inosine,uridine,deoxyguanosine,cytidine monophosphate,uridine monophosphate,guanosine monophosphate and adenosine monophosphate in Wuling Capsules/Wuling fungus powder.METHODS The analysis of aqueous extract of this drug was performed on a 40℃thermostatic Thermo Hypercard column(2.1 mm×100 mm,5μm),with the mobile phase comprising of water(containing 2 mmol/L ammonium acetate,0.06%ethylenediamine)-acetonitrile(containing 2 mmol/L ammonium acetate,0.06%ethylenediamine)flowing at 0.6 mL/min in a gradient elution manner,and electron spray ionization source was adopted in negative ion scanning with multiple reaction monitoring mode.RESULTS Ten constituents showed good linear relationships within their own ranges(R^(2)>0.9900),whose average recoveries were 85.52%-104.21%with the RSDs of 0.79%-4.48%.CONCLUSION This simple,feasible and reproducible method can be used for the quality control of Wuling Capsules/Wuling fungus powder.
