摘要
目的研究小花清风藤Sabia parviflora Wall.ex Roxb.叶化学成分及其与JAK2/STAT3蛋白的结合活性。方法小花清风藤叶70%乙醇提取物采用硅胶、Sephadex LH-20、ODS反相硅胶、半制备HPLC进行分离纯化,根据理化性质及波谱数据鉴定所得化合物的结构。采用AutoDock Vina软件对化合物与JAK2/STAT3蛋白分子进行对接。结果从中分离得到8个化合物,分别鉴定为木犀草素(1)、山柰酚(2)、(2,5-dioxo-4-imidazolidinyl)-carbamic acid(3)、grossamide(4)、柴胡皂苷A(5)、柴胡皂苷D(6)、柴胡皂苷B2(7)、柴胡皂苷B1(8)。分子对接结果表明,山柰酚及木犀草素分别对JAK2蛋白和STAT3蛋白具有较好的亲和力。结论化合物3为首次从清风藤属植物中分离得到,化合物1~2对JAK2/STAT3蛋白具有潜在的靶点结合活性。
AIM To study the chemical constituents from the leaves of Sabia parviflora Wall.ex Roxb.and their binding activities with JAK2/STAT3 proteins.METHODS The 70%ethanol extract from the leaves of S.parviflora was isolated and purified by silica gel,Sephadex LH-20,ODS reversed-phase silica gel and semi-preparative HPLC,then the structures of obtained compounds were identified by physicochemical properties and spectral data.Autodock Vina software was used to dock the compounds with JAK2/STAT3 protein molecules.RESULTS Eight compounds were isolated and identified as luteolin(1),kaempferol(2),(2,5-dioxo-4-imidazolidinyl)-carbamic acid(3),grossamide(4),sailkosaponin A(5),sailkosaponin D(6),sailkosaponin B2(7),sailkosaponin B1(8).Molecular docking result showed that kaempferol and luteolin had good affinities for JAK2 protein and STAT3 protein,respectively.CONCLUSION Compound 3 is isolated from the genus Sabia for the first time.Compounds 1-2 have potential target binding activities with JAK2/STAT3 proteins.
