膝痹康颗粒中挥发油和有效成分的提取工艺研究

摘要 目的优化膝痹康颗粒的最佳提取工艺。方法以膝痹康标准方剂为参比,以挥发油、地黄苷D、益母草苷、毛蕊花糖苷、总多糖、阿魏酸、Z-藁本内酯、浸膏得率为评价指标。在挥发油提取优化方面,用正交实验考察加水量、浸泡时间、提取时间3个因素对挥发油得量的影响;在活性成分提取优化方面,用正交实验考察乙醇体积分数、溶剂倍数、提取时间对提取效果的影响,确定最佳提取工艺。结果提取时间是影响挥发油提取量的关键因素(P<0.05),挥发油的最佳提取条件为用8倍水浸泡30 min、提取9 h。在提取完挥发油后,再进行乙醇回流提取,乙醇体积分数、溶剂倍数、提取时间均为影响成分提取的关键因素(P<0.05),地黄苷D、益母草苷、毛蕊花糖苷、总多糖、阿魏酸、Z-藁本内酯、浸膏得率的最佳提取条件为加入7倍体积分数为70%的乙醇回流提取2 h。挥发油、地黄苷D、益母草苷、毛蕊花糖苷、总多糖、阿魏酸、Z-藁本内酯、浸膏得率的RSD值分别为1.34%、1.92%、1.16%、2.68%、2.51%、1.18%、1.20%、3.08%,验证实验表明该工艺稳定,可获得稳定的挥发油和冻干粉末颗粒。结论该方法稳定可行,可作为膝痹康颗粒的提取工艺。 Objective To optimize the extraction process of Xibikang Granules.Methods By taking the standard prescription of Xibikang as reference,the yields of volatile oil,rehmannioside D,leonuride,verbascoside,total polysaccharide,ferulic acid,Z-ligustilide and extract rate were taken as the evaluation indexes.For volatile oil extraction,orthogonal test was used to investigate the influence of water addition,soaking time and extraction time on the yield of volatile oil.For active ingredient extraction,orthogonal test was used to investigate the influence of ethanol volume fraction,solvent volume and extraction time on the extraction effect.Results The extraction time was the key factor affecting the extraction amount of volatile oil(P<0.05).The optimal extraction condition of volatile oil was 8 times water immersion for 30 min and extraction for 9 hours.After the volatile oil extraction completed,ethanol reflux extraction was carried out.Ethanol volume fraction,solvent volume and extraction time were the key factors affecting the extraction of components(P<0.05).The optimal extraction conditions of rehmannioside D,leonuride,verbascoside,total polysaccharide,ferulic acid,Z-ligustilide and extract rate were as follows:adding 7 times volume of 70%ethanol for reflux extraction for 2 hours.The RSD of volatile oil,rehmannioside D,leonuride,verbascoside,total polysaccharide,ferulic acid,Z-ligustilide and extract rate were 1.34%,1.92%,1.16%,2.68%,2.51%,1.18%,1.20%,and 3.08%,respectively.The test showed that the process was stable and stable volatile oil and lyophilized powder particles could be obtained.Conclusion The method is stable and feasible,and can be used as the extraction process of Xibikang Granules.
