蠲毒消瘤饮抑制结肠癌细胞的作用机理

Mechanism of Juandu Xiaoliu Yin inhibiting colon cancer cells

目的探讨蠲毒消瘤饮对结肠癌细胞增殖、迁移和侵袭的作用及机制。方法以人结肠癌HCT116细胞为模型,分别使用平板克隆形成实验,对细胞的克隆形成能力进行增殖检测;运用Transwell肿瘤细胞侵袭实验和细胞划痕运动实验,分别检测蠲毒消瘤饮对HCT116细胞侵袭和迁移的影响;采用Western blot实验检测蠲毒消瘤饮对VEGF/MEK/Ras/Raf信号通路上关键蛋白表达。结果蠲毒消瘤饮作用后,HCT116细胞增殖、迁移和侵袭受到显著抑制,细胞内VEGF/MEK/Ras/Raf蛋白表达水平显著降低。结论蠲毒消瘤饮在体内体外具有抑制结肠癌细胞生长的作用。

Objective To investigate the effect and mechanism of Juandu Xiaoliu Yin on the proliferation,migration and invasion of colon cancer cells.Methods Human colon cancer HCT116 cells were used as model to detect the proliferation of clonogenesis ability by plate cloning assay.Transwell tumor cell invasion assay and cell scratch exercise assay were used to detect the effects of Juandu Xiaoliu Yin on the invasion and migration of HCT116 cells.Western blot assay was used to detect the effect of Juandu Xiaoliu Yin on the expression of key proteins in VEGF/MEK/Ras/Raf signaling pathway.Results The proliferation,migration and invasion of HCT116 cells were significantly inhibited,and the expression levels of VEGF/MEK/Ras/Raf protein were significantly decreased after treatment with Juandu Xiaoliu Yin.Conclusion The traditional Chinese medicine Juandu Xiaoliu Yin can inhibit the growth of colon cancer cells in vitro and in vivo.
