摘要 为探究PRP1基因与云上黑山羊产羔数性状的关联及其在山羊卵巢组织中的表达,试验以高产羔数和低产羔数云上黑山羊为研究对象,采用PCR克隆、Sanger测序和生物信息学分析解析PRP1基因与云上黑山羊产羔数性状的关联。PCR结果表明,云上黑山羊PRP1基因CDS区长度717 bp,编码238个氨基酸。Sanger测序结果表明,PRP1基因存在g.1821248 T>C和g.1821220 A>G两个位点,这两个突变位点均未造成氨基酸编码蛋白数量的改变;PRP1二级结构主要由无规则卷曲和延伸链构成,最小结构自由能和热力学系的自由能发生改变;蛋白质三级结构的空间构型未发生改变。PRP1蛋白为亲水性蛋白,不存在蛋白结构域;蛋白互作预测表明了PRP1蛋白具有与其他功能蛋白相互作用的能力。 The aim was to investigate the association of PRP1 gene with litter size and its expression in ovary tissue in Yunshang Black goats.In this paper,the association of PRP1 gene with different litter sizes in Yunshang Black goats were studied by PCR,Sanger sequencing and bioinformatics analysis.The PCR results showed that the CDS region of the PRP1 gene in Yunshang Black goats was 717 bp in the protein expressed by the gene was length and the protein expressed by the gene was encoded by 238 amino acids.Sanger sequencing results showed that there were two loci,g.1821248 T>C and g.1821220 A>G,in the PRP1 gene,and neither of these two mutated loci have not caused any change in the amount of amino acid-encoded protein.The secondary structure of PRP1 consists mainly of irregularly coiled and extended chains with altered minimum structural free energy and free energy of the thermodynamic tether;the spatial configuration of the tertiary structure of the protein is unaltered.This study also revealed that the PRP1 protein was hydrophilic and had no protein structural domain;the protein interaction prediction indicated that the PRP1 protein had the ability to interact with other functional proteins.
