摘要
为探讨小尾寒羊不同肌肉组织中MSTN基因的表达差异,根据GenBank已发表序列(NM-001009428.1)设计1对引物,以小尾寒羊肌肉组织总RNA为模板,采用实时荧光定量PCR技术(qRT-PCR)分析了MSTN基因在公羊和母羊前腿肌肉、后腿肌肉、腹部肌肉、背最长肌和后腱肉中的表达量。结果表明,MSTN基因在小尾寒羊公羊背最长肌中的表达量最高,然后依次是后腱肉、前腿肌肉和腹部肌肉,后腿肌肉中表达量最低;在母羊后腿肌肉中的表达量最高,然后依次是后腱肉、背最长肌和腹部肌肉,前腿肌肉中的表达量最低。MSTN基因在公羊背最长肌中的表达量极显著高于母羊(P<0.01),在后腿肌肉中的表达量极显著低于母羊(P<0.01),后腱肉、前腿肌肉和腹部肌肉公羊与母羊之间差异不显著(P>0.05)。说明MSTN基因在小尾寒羊不同肌肉组织中的表达量存在差异,在公、母羊个体之间的表达量也存在差异,其差异可能会影响产肉性能。
To study the expression of MSTN gene in different muscle tissues of Small-tailed Han sheep,a pair of primers were designed according to the published sequence of GenBank(NM-001009428.1)in this study.Using the total RNA of muscle tissue of Small-tailed Han sheep as template,real-time quantitative PCR(qRT-PCR)was used to analyze the MSTN gene expression levels of foreleg muscle,hind leg muscle,abdominal muscle,longissimus dorsi muscle and posterior tendon meat in male and female sheep.The results showed that the expression of MSTN gene was the highest in longissimus dorsi muscle of Small-tailed Han sheep male sheep,followed by the muscle of posterior tendon,foreleg and abdomen,and the lowest in the muscle of hind leg.The expression level was the highest in the hind leg muscle of female sheep,followed by the posterior tendinous muscle,longissimus dorsi muscle and abdominal muscle,and the lowest in the foreleg muscle.The expression level of MSTN gene in longissimus dorsi muscle of male sheep was significantly higher than that of ewes(P<0.01),but the expression level in hindleg muscle was significantly lower than that of female sheep(P<0.01),and there was no significant difference between male sheep and female sheep in posterior tendon meat,foreleg muscle and abdominal muscle(P>0.05).These results indicate that there are differences in the expression level of MSTN gene in different muscle tissues of Small-taied Han sheep,and there are also differences in the expression level between male and female sheep,which may affect the meat production performance.
